The last two decades have seen a revolutionary change in the dogma suggesting that the CNS operates optimally without any assistance from circulating immune cells. Previously, attempts were made to globally mitigate any immune activity in the brain, assuming such activity was a sign of ongoing pathology. We now know that a continuous dialogue occurs between the brain and the circulation, and that soluble and cellular components derived from the circulation support brain plasticity. The implications of these finding for brain aging and neurodegenerative diseases are becoming critical issues in finding cures for these conditions. Moreover, the mechanisms whereby the brain controls the activity of the immune system is an additional mystery in this bi-directional relationship. The Advanced Course will feature contributions by scientists who have made fundamental contributions to understanding this evolving new field.

Structural biology is entering an exciting new phase. Over 10,000 new structures are being determined every year. Powerful new cryo-EM and X-ray laser approaches promise to vastly improve the ease of structure determination for large molecules and complexes and to provide a time-resolved view of molecular machines at work. With these tools and biochemical assays, ever more rapid progress is being made in understanding key biological processes and how they can be manipulated. This symposium will bring together students and senior investigators developing new methods for structural analysis, determining new structures, and using structures to understand biology. The symposium will encourage and facilitate discussion and interactions between junior and senior scientists. The symposium will feature about 40 talks and 100 poster presentations.

Accumulating evidence points to brain reorganization with chronic pain. It seems that the grey matter of the neocortex dynamically changes with chronic pain and this reorganization is pain type specific. In parallel to the anatomical changes, brain functional properties change as well, and the best evidence for condition type functional reorganization is now documented in resting state brain activity, where one can identify a core set of brain networks commonly disturbed with chronic pain, as well as condition specific functional properties. Moreover, brain functional network properties provide a signature for existence and for emergence of chronic pain both in humans as well as in rodent models for pain chronification. It is common clinical knowledge that although a very large patient population presents with similar injuries that give rise to pain, only a small minority of them develop chronic pain. Thus the critical question in the field of pain research is: what characteristics differentiate between those that develop chronic pain and the ones who properly recover from their injury into health. Complimenting the human study there is now evidence in animal models regarding brain reorganization with pain chronification.

Single cell omics is a new frontier in biology, where genome-wide data is captured at the resolution of individual cells. Single cell high-throughput technologies are being developed in the areas of imaging, mass spectrometry and DNA- and RNA-sequencing, and combinations thereof. The corresponding software and visualization tools are also a large active field of research. Single cell omics has important implications for all areas of development, differentiation and cellular decision-making, because these can now be studied at unprecedented resolution. Equally, single cell omics methods are shedding new light on immunity and cancer, due to the heterogeneous cell populations involved in both areas of biology. Therefore, these approaches are revolutionizing both basic biology as well as our understanding of disease, and technologies involved in translation to medicine. Since the field is in its infancy, and there are few meetings dedicated to this topic, there is a huge unmet need in the community to congregate and exchange ideas and information about new methods and the biological insights gained. A meeting such as this will be of continuing importance until single cell omics technologies become commonplace and established, and integrated into all areas of biology.

Recent ground-breaking developments in neuroscience, such as optogenetics, in vivo 2-photon confocal microscopy, head mounted microscopes, powerful new developments in modeling, behavioral neuroscience approaches, and sophisticated brain imaging tools, have changed dramatically studies of memory. Most importantly, these developments have fostered interdisciplinary studies that led to integrated molecular, cellular, systems, cognitive and behavioral explanations of how memories are allocated, formed, consolidated, reconsolidated and retrieved. These studies have also led to mechanistic cross-disciplinary studies of memory disorders, which in some cases led to the development of targeted treatments that are changing how we imagine treating the considerable health burden associated with this large class of conditions.

Cell death has long been known to be an important instigator of inflammation in sterile injury, as well as an amplifying factor in infection-associated inflammation, but the specific molecules underpinning this remain obscure. Understanding how dead and dying cells initiate and escalate inflammation has important implications for our understanding and treatment of autoinflammatory and infectious diseases. This meeting will explore current knowledge concerning the cellular constituents that drive inflammation (i.e., damage-associated molecular patterns), as well as how abnormal modes of cell death, such as necrosis, necroptosis and NETosis, can perturb inflammatory outputs and responses to dying cells. In particular, the conference aims to explore gaps in our current understanding of how dead and dying cells influence inflammatory responses in disease settings and will bring together experts in two major research fields, ”cell death” and ”inflammation,” for the purpose of clarifying what the key questions and therapeutic targets are in this rapidly evolving area. Many well-known physiological drivers of cell death (e.g., TNF, TRAIL, CD95/Fas), as well as the molecules that transduce signals from these receptors (e.g., RIPKs, IAPs, TRAFs, IKKs), are also centrally involved in promoting inflammation. However, the tremendous overlap between cell death and inflammatory signaling is only becoming appreciated of late, and although it is also widely accepted that the constituents of healthy cells can drive inflammation upon release into the extracellular space, the identity of these cellular constituents is still a matter of debate. This meeting will focus on teasing out the relationships between cell death and inflammatory signaling to highlight how perturbation of either of these processes invariably impacts upon the other. This meeting will also explore the nature of the molecules that promote and modulate inflammation during cell death. The meeting will bring together scientists from diverse fields (cell death, inflammation, cancer, innate immunity) that would not normally interact, and will identify and explore the key questions and directions that will shape the future of research in this field.

The epitranscriptome is a new emerging field of research. There will also be a medical perspective, discussing how the lack of certain modifications leading to aberrant innate immune recognition of nucleic acids results in disease.

Steroid hormone action impacts many aspects of physiology and disease states. Although scientists in the field traditionally focused on how nuclear steroid receptors mediate gene transcription, it is now established that these same receptors exist and function at various cellular locations including the cell membrane and mitochondria. Thus this conference focuses on the latest advances and approaches to understand how signaling from extra-nuclear and nuclear steroid receptors integrate to produce genomic and non-genomic biological effects. This year's SRC focuses on steroid hormone action in cancer, metabolism, neuroscience, cardiovascular effects, ion channels, lung disease. Many new speakers will give talks and the opportunity for trainees to present their work in oral and poster sessions will be emphasized as well. Most importantly, many productive collaborations between scientists, junior and senior, have occurred from the interactions during the meeting. Junior scientists and trainees are especially encouraged to interact with senior faculty, in multiple venues that encourage this during the meeting. Travel funds for trainees are also a priority of this meeting.

The overarching goal of the meeting is to bring together as many scientists working on diverse aspects of GRK/arrestin signaling as possible, in order to facilitate the exchange of novel findings and to stimulate the generation of new hypotheses, collaborations and approaches. The structural and functional mechanisms of GRKs and arrestins, as well as their roles in biased/non-canonical signaling by GPCRs, in the actions of drugs of abuse, in cardiovascular disease and in cancer will be major highlights of the meeting. Sessions devoted to these topics will foster further studies to combat addiction and disease with the ultimate objective of positively influencing human health. There are many opportunities available for attendees to give oral and poster presentations as well as optional recreational activities to foster the exchange of ideas and formulate new collaborations.

This SRC is focused on protein aggregation, which plays central roles in health and in disease and a converged understanding of these dual roles is absolutely essential if we are to understand the emergence of biological complexity and the onset and progression of devastating systemic and neurodegenerative diseases. Importantly, protein aggregation and aggregates are also central in numerous biological functions as is evidenced in the growing number of accounts of functional aggregates and their roles in fungi, bacteria, and in the innate immune system of humans. Recent studies have highlighted the role of phase separation driven by a form of protein aggregation that gives rise to functional, non-stoichiometric assemblies, known as membraneless organelles. The cross-disciplinary nature of the conference is accentuated by the program, which brings together biophysicists, structural biologists, and bioengineers with cell biologists and clinical scientists.

This SRC focuses on the endoplasmic reticulum (ER) and its functions. The purpose of the meeting is to merge recent developments in molecular biology, cell biology, genetics, and physiology aimed at unraveling the complexities of secretory protein folding, ER stress pathways, lipid signaling, and ER membrane biology, and how failure of these processes lead to pathologies and disease. This SRC will include talks from outstanding scientists, both invited and selected from poster abstracts. The meeting is known for its active discussions, not only during the presentation session, but also during the free afternoons, and especially during the poster sessions. Posters will remain on display during most of the conference and a selection will be advertised in oral poster pitches. The venue, Vermont Academy, is excellently suited for the mingling of junior and established scientists, which will be strengthened by meet-the-speaker sessions that allow discussions of both career issues and science.

This Science Research Conference will focus on the most recent advances in the fields of signal transduction in the immune system, with an emphasis on the coupling of signaling to differentiation pathways, the development of new technologies for understanding and imaging signaling, interactions between metabolism and lymphocyte activation and differentiation, signaling through mechanotransduction, and signaling through immunomodulatory receptors in cancer. These topics have direct relevance to a number of clinically important areas, including autoimmunity, immunodeficiency diseases, transplantation immunology, tumor immunology, infectious diseases and chronic inflammation. In particular, leukocyte metabolism, the mechanism and feedback of antigen receptor signaling initiation and kinase regulation, immunomodulatory receptors, and cytoskeletal changes that accompany leukocyte activation and migration have provided a wealth of new information that will enhance our understanding of the complex pathways that control immune responses under normal as well as pathogenic situations. As such, the mechanisms and pathways that will be discussed in detail at this conference represent a major source of target leads for the development of new therapeutics to treat immunological disorders and sit at the forefront and intersection of basic and translational research. The inclusion of sessions that bridge these two types of research is another invaluable contribution of this meeting.There will be a large number of selected talks from the abstracts, which will give junior investigators an opportunity to present their data.

This SRC is focused on the dynamic nature of mobile DNAs in mammalian genomes and their phenotypic impacts. Topics will encompass mechanistic and evolutionary aspects of diverse types of retrotransposons, including LINE-1 and endogenous retroviruses, as well as DNA transposons and related human genes. Attendees will also explore interactions between these sequences and the epigenome and endogenous machinery of DNA repair and rearrangement. These elements comprise are a major portion of our DNA, and are significant contributors to genomic diversity in the form of structural variation. While the germ line is a well-known site of activity of mobile genetic elements, there is increasing evidence for expression and mobility in somatic cells and in human diseases, particularly cancers. This makes it an exciting time for the field. Moreover, new insights are being gained into roles of mobile elements in shaping gene regulatory networks in normal cells. Finally, harnessing mobile genetics elements as tools for forward genetic screens or gene delivery has provided new capabilities to change the genome. This is a long standing meeting, certain to be thought provoking, animated, and collegial all at once.

This SRC will focus on the process of transcription in prokaryotes, from structure/function investigations to transcription networks and system level regulation. In all organisms, controlling the process of transcription is fundamental to gene expression, gene regulation, and development. RNA polymerase, which transcribes genomic DNA into RNA, is the complex protein machine that performs transcription. Additional factors that alter initiation, elongation, or termination of transcription program an amazing variety of developmental pathways. Biochemical and genetic investigations of transcription regulation have a long history of revealing the myriad ways that gene expression can be controlled. In recent years this work has been augmented by the availability of atomic-level structures of prokaryotic and eukaryotic RNA polymerases and transcription factors in combination with systems-wide profiling of gene regulatory events. Together this work has revealed that many similar structure-function relationships underlie the workings of the transcriptional machinery from the simplest bacterium to a human cell. Attendees can expect exciting sessions on RNA polymerase structural biology, mechanisms that control initiation, elongation, and termination, regulation of transcription in pathogens, RNA-based control, transcriptional networks, and connections between cell topology and gene expression.

The maintenance of genome integrity is critical for the suppression of cancer and premature ageing. Only recently has it become appreciated that DNA replication stress is a crucial driver of genomic instability. The timely progression of replisomes can be disrupted by lesions and secondary structures in the template, by bound proteins and by conflicts with the transcription machinery. A prolonged pause of the replisome then exposes single stranded DNA, which, due to its recombinogenic nature, can lead to genome rearrangements, fragile site expression and cell death. Importantly, some cancers present excessive endogenous levels of replication stress, which can be exploited for their clearance. In this conference, we aim to bring together scientists studying DNA replication and repair, with those interested in how DNA damage can influence cancer and ageing.

Understanding immune tolerance and its breakdown is necessary to treat autoimmune diseases such as lupus, multiple sclerosis, uveitis, type 1 diabetes, arthritis, inflammatory bowel disease, and many others. We bring together basic scientists with clinicians and industry experts to present and discuss new insights into the triggers, pathogenesis and therapy of chronic inflammatory and autoimmune disease. Eight major sessions will cover a broad range of topics in development, pathogenesis and therapy of autoimmune disease, starting with the effects of genetics and environment, through the role of innate and adaptive immune elements as well as the tissue itself, the role of commensal microbiome and metabolism, and finally translational research and clinical approaches. Each thematic session will feature 4 invited speakers and two talks selected from submitted abstracts. Participants have ample opportunity for interaction and discussion. Spontaneous mingling of junior and established scientists will be strengthened by meet-the-speaker sessions that allow discussions of both career issues and science. Since 1994, this Biennial FASEB Science Research Conference has been devoted to exciting and groundbreaking research on viral, bacterial, fungal, and protozoal pathogens. The combined themes are particularly pertinent as we now know that regardless of their taxonomic differences, microbes confront similar barriers from the host innate and acquired immune systems, thus indicating both similar and unique virulence strategies.As such, this is the only long-standing specialized international conference that regularly attracts virologists, bacteriologists, mycologists, parasitologists, immunologists, antimicrobial researchers, and translational scientists. Notable speakers include – but are certainly not limited to – Gordon Dougan, Kim Orth, Vern Carruthers, Erica Ollman Saphire, Gerry Wright, Vanessa Sperandio, Russell Vance, and many others. By convening together leading scientists with expertise in all areas of infectious disease, this conference optimizes information transfer, catalyzes provocative discussions, and fosters long-term cross disciplinary collaborations. In 2017, the program will include sessions on diverse aspects of host-microbe interactions from molecular, evolutionary, immunological, and therapeutic perspectives. Talks will also include new, state-of-the-art approaches as well as how to adapt traditional approaches to meet today’s challenges. NAD and its metabolites are important regulators of cancer, metabolic disease, autoimmunity, and aging. They play important roles in energy metabolism, function as signaling molecules, and as protein modifications such as ADP-ribosylation. This diversity of function demonstrates the biological importance of NAD and the interconnectivity between cellular metabolism, physiology, and cellular signaling. Recently, significant progress has been made in our understanding of NAD that highlight its broad relevance to human health and disease. This SRC brings together all of the leading experts in NAD biology to one venue with the goal of advancing our understanding of the molecule. This SRC will highlight the latest research on mechanisms of ion channel regulation and its impact on the physiology of diverse cell types. A major goal of this conference is to stimulate discourse, seed new ideas, and facilitate collaboration in ways that will accelerate new discoveries about ion channel regulation at both the basic and translational levels. This conference aims to fuel the ‚engine of discovery‘ by bringing together scientists applying all of these diverse technical and conceptual strategies to study ion channel regulation across a range of different biological systems.  The program consists of nine scientific sessions and a keynote address, as well as two breakout sessions and lunches focused on trainee career development. Scientific sessions are organized around themes, rather than ion channel subtype, to foster crosstalk between fields, and will cover a variety of topics including: sensory systems and pain; regulation of neuronal excitability; regulation of gene expression; ion channels in disease; ion channel structure and drug development; recent advances in imaging and ion channel cell biology; regulation of the cardiovascular system and skeletal muscle; regulation of ER Ca2 stores and other organelles; celebrating Bill Catterall and ion channels.  Presentations in these sessions will be made by both new, up-and-coming investigators as well as established leaders in the ion channel field, including several members of the National Academy of Sciences. Each session will also include two short-talks to be selected from the abstracts, with an emphasis on presentations by graduate students, postdoctoral trainees, and junior faculty.

This SRC presents progress in research on diverse aspects of genetic recombination, a critical process that maintains integrity of the genome and that ensures the faithful transmission of the genome between generations. This conference focuses on the study of recombination mechanisms, the impact of recombination on genome integrity, and the influence of other DNA processes on recombination. Many of the recent advances in understanding recombination in normal, pathological, and environmentally stressed contexts have emerged through new combinations of experimental approaches spanning biochemistry, biophysics, genetics, genomics, and cytology. The agenda reflects the diversity of approaches currently used and is specifically geared toward identifying the emerging technologies and concepts that are moving the field forward. The meeting will focus on new, unpublished data in the field and will highlight connections between recombination and other cellular processes, including transcription, replication, cell cycle progression, and other branches of DNA repair. There will be presentations by researchers using diverse model organisms including bacteria, fungi, flies, nematodes, and mammalian cells, as well as in vitro biochemistry and structural biology. There will be many opportunities for interaction between established investigators and scientists who are new to the field, and a significant number of the plenary speakers will be chosen from submitted abstracts.

This SRC will bring together international scientists from academia and industry for lively discussions on the latest developments in the growth hormone (GH)/prolactin (PRL) family of hormones and their clinical applications. Beyond their classic roles in growth and lactation, these related hormones are increasingly recognized regulators of metabolism, stem cells, aging, behavior, cancer, and perinatal health of both mother and child, via actions on diverse target tissues. Moreover, they influence major associated health problems, such as obesity, diabetes, aging, mental health and multiple cancers. Our goal is to bring together senior and junior investigators working on all aspects of these hormones, from cell biology, genetics, developmental biology, neurobiology, metabolism, physiology, to their clinical and agricultural applications. Together, we will increase our understanding of how GH/PRL family proteins modulate health and disease processes, forge new collaborations, and accelerate the discovery of new pharmacologic approaches. A major feature of life sciences in the 21st century is their transformation from phenomenological and descriptive disciplines to quantitative and predictive ones. Revolutionary opportunities have emerged for mathematically driven advances in biological research. However, the emergence of excessive complexity in self-organizing biological systems poses fundamental challenges to their quantitative description. Dealing with the connectivity and transformation of different components in a space, topology provides a dramatic simplification of biomolecular data and sheds light on drug design, protein folding, organelle function, signaling, gene regulation networks and topology-function relationship.

This conference focuses on the biology of protein kinases and phosphorylation signaling. Among FASEB’s longest-running SRCs, it brings together a dynamic international community of junior investigators, trainees, and established researchers from a field that is achieving startling new insight and medical breakthroughs. The program will link detailed understanding of protein structure and biochemical mechanism to control of large-scale processes like the cell division cycle, cell morphogenesis, and metabolism. This SRC will explore the field’s latest research, which is revealing surprising mechanisms of regulation, previously unappreciated subcellular organization, logic of pathways that coordinate diverse intracellular systems. Importantly, derangement of kinase signaling underlies disease, including cancer and diabetes. July 17, 2017 is the date that marks the 100th anniversary of the birth of Full Member of the USSR Academy of Sciences Dmitri K. Belyaev (1917–1985), an outstanding scientist, evolutionist and geneticist. In view of this memorable date, the Institute of Cytology and Genetics of the Siberian Branch of the Russian Academy of Sciences and the co-organizers will be holding Belyaev Conference. Belyaev authored an all-new chapter in the theory of morphogenetic processes that explores interactions between two main factors in evolution: variability and selection. In an attempt to find the answers, he launched a large-scale, still ongoing experiment aimed at reproducing the earliest stage of the animal domestication process, with the silver fox as the test subject. The key was selection for human-friendly responses in the fox. As a result, a unique, world-famous tame fox population was created.

Many cellular constituents in the human brain permanently exit from the cell cycle during pre- or early postnatal development, but little is known about epigenetic regulation of neuronal and glial epigenomes during maturation and aging, including changes in cognitive and psychiatric disease. Normal brain development and function is dependent on highly regulated mechanisms governing DNA cytosine methylation and hydroxymethylation, and probably more than 100 residue-specific histone modifications associated with gene expression and silencing and various other functional chromatin states. Equally important is the 3-dimensional organization of the chromosomal material inside the cell nucleus, commonly referred to as the ‘3D Genome’, with chromosomal loopings potentially bypassing hundreds of kilobases on the linear genome to enable promoter-enhancer interactions and other mechanisms important for transcriptional regulation. Combined exploration of epigenomic and 3D genome chromosomal conformation maps maps is likely to illuminate the role of regulatory non-coding sequences in neuropsychiatric disease.

This is the leading international conference in lysophospholipid biology, attracting participants from all continents. The Course will start with critical analysis of the evolution of the study of various forms of long-term potentiation and long-term depression and what is generally accepted about their basic mechanisms, including a discussion of modern approaches to understanding the mechanisms and functions of synaptic plasticity. A wide range of areas will be covered, including: (a) the molecular mechanisms underlying different forms of short-term synaptic facilitation and depression that last no more than a few minutes and are typically mediated by presynaptic mechanisms that alter neurotransmitter release (b) the function of key postsynaptic proteins found in the postsynaptic density as well as the role of neurotransmitter receptor regulation during long-term synaptic and experience-dependent plasticity (c) the methodological approaches that have been recently developed to measure the dynamic localization of synaptic elements at nanoscale resolution and how these nanoscale morphological and functional properties regulate synapses (d) the role of neurogenesis throughout life as a crucial mechanism underlying learning-induced circuits in the dentate gyrus and the olfactory bulb (e) how these forms of plasticity integrate to result in the restructuring of entire neuronal circuits. Participation in the Advanced Course will provide an essential conceptual and methodological framework for anyone intending to pursue rigorous research. The advantages and disadvantages of each method, preparation and recording technique are considered in relation to the specific scientific questions being asked. The Advanced Course will not only develop participants’ skill but will also provide plenty of time for informal gatherings with some of the world leaders in synaptic plasticity, designed to foster brainstorming across different disciplines.

Age is the greatest risk factor for multiple neurodegenerative diseases. In that context, the prevailing perception of the aging brain is of linear decline across multiple systems. A diametrically different perspective is that the aging brain is a dynamic adaptive organ that initiates multiple survival systems throughout the aging process. While these adaptations in the aging brain are for the majority of persons beneficial, they can, in vulnerable populations, lead to increased risk of neurodegenerative disease. The systems biology of transition states in normal aging and neurodegeneration provides insights into mechanisms of disease risk and disease progression. Risk of multiple neurodegenerative diseases can emerge during neuroendocrine transitions of aging when gene networks undergo shifts in activation and expression. These risk networks underlie the prodromal phase of neurodegenerative disease. Later, transition states typify the progressive nature of multiple neurodegenerative diseases. Risk for and progression of Alzheimer’s disease are exemplars of both risk that can emerge during neuroendocrine transitions and the transition states of disease progression.

Translational control is a major focus of attention that extends to many fields, including developmental biology, neurobiology, cell physiology, disease, synthetic and systems biology, etc. In addition to translation of messenger RNA, the conference will feature advances in the field of non-coding RNAs, both small (miRNA) and large (lncRNA), that influence the translation process and its machinery. Novel, powerful high-throughput technologies and structural advances will also be central, as they are pushing the field forward in manners that were unthinkable only few years ago, and are helping to create a “systems” view of translation.

With the increasing appreciation of the integration between different steps of gene expression, the conference will explore the connections between translation, mRNA turnover, nonsense-mediated decay, RNA localisation, and nuclear events such as transcription and splicing. Finally, many alternatives to the classical “one message makes one protein” concept are being unraveled which increase the potential for regulation. These alternatives will be featured, as well as the role of translational regulation in diseases as diverse as cancer, neurological disorders or infectious diseases.

Neuroscience has entered a critical phase, with several international large-scale efforts devoted to the analysis and understanding of brain function. Studies exploiting new technologies for defining the connections in the brain, driving neuronal activity and modeling of neuronal processing are each hampered by our incomplete knowledge of the brain’s constituent parts. Fortunately, a bevy of single cell analyses are now being deployed to better understand the cells that constitute the functioning nervous system. These approaches offer insights into how cells function individually as the building blocks for understanding how they work in ensemble to create functional pathways. A set of emerging tools are available to assess the molecular and cell biology of single cells, including genomic analyses of somatic mutations, epigenomics to explain the influence of the environment upon genome expression, and transcriptomics to gain insight into the capacity of a cell to synthesize proteins and modulate its environment. Powerful technologies permit a variety of cell constituents to be addressed, including proteins, post-translational modifications, peptides and metabolites. Novel biosensors offer parallel analyses of cellular physiological states, reporting on the pH, membrane voltage and metabolic state. Multiplexed together, such methodologies offer a far more complete biological understanding than the more typical gene expression analyses, providing a rich picture of each single cell’s biology in their normal context – information needed to fully interpret the meaning of results from transcriptomics and proteomics studies.This Advanced Course is designed to provide the foundational knowledge needed to critically evaluate the design and execution of single cell omic studies, through daily lectures by the Faculty, critical reading of papers and extensive didactic discussion sessions. In the sessions, discussion will focus on the proper framing of scientific questions to best utilize these techniques, the limitations and advantages of each technique, and what the data can tell us about neuronal function.
