Understanding how mitochondria communicate within the cell will provide important clues to elucidate its roles in normal cell physiology, as well as numerous diseases such as diabetes, neurodegeneration and cancer. The classical view of mitochondria is of an organelle that interacts with other features of cell biology primarily through the provision and consumption of metabolic intermediates and energetic products. Emerging evidence from many areas of science is accumulating to suggest that mitochondria play much more active roles in communication with other organelles, determining cell and organismal behavior. This conference will focus on these integrated behaviors and the resulting communication. The sessions will be organized around the different nodes, modes and destinations of that signaling. It will bring together people and ideas from different areas of cell biology and physiology that typically do not attend the same conferences, resulting in the development of new collaborations and concepts.

The overall goal for this meeting is to bring together scientific leaders and trainees with a common interest in PI3Ks and relevant downstream physiological processes. It will address advances in basic molecular and cellular mechanisms that are regulated by PI3Ks, other PI kinases and, their cognate phosphatases; highlight new advances in the characterization of PI3K pathway alterations in genetic syndromes that affect growth and the immune and nervous systems; and provide an important update on the clinical development of PI3K pathway inhibitors, and implications for the burgeoning field of cancer immunotherapy. Whole exome sequencing studies have increasingly implicated PI3K signaling in diseases (immune, growth and neurological disorders) which may benefit from drugs currently in development for cancer and inflammation. This meeting will be an important forum for discussing approaches to treating these additional disorders.

Carcinogenesis is the result of a complex interplay of cell intrinsic and extrinsic processes that promote genomic instability, sustained proliferation, resistance to apoptosis, reprogramming and reorganization of the stromal environment. An immune cell infiltrate is a characteristic feature of many tumors, and it is increasingly appreciated that immunity and inflammation are key determinants of tumor development and progression. This meeting will consider the molecular and environmental factors that shape the extensive cross-talk between immune, stromal and cancer cells in the tumor microenvironment. Positioned at the interface of cancer cell signaling, stem cells, inflammatory pathways and microbial drivers, this meeting should provide new insights into the factors that control the balance between tumor-promoting and tumor-suppressive immunity and inflammation and how this can be applied in the prevention and treatment of cancer. The meeting will attract a multi-disciplinary group of cancer cell biologists, immunologists and inflammation biologists and will be of interest to basic and clinical scientists alike.

Autophagy is a quality control mechanism that plays an important role in health and disease, including neurodegeneration, cancer, infection and inflammation. The core autophagy machinery has been identified and studied in detail. In addition, integration and cross-talk with other cellular pathways is becoming clearer, providing a rationale for why autophagy is linked to so many diverse physiological functions. Future challenges are focused on integrative structural, molecular and system approaches that are able to provide a much more refined understanding of autophagy regulatory networks. This meeting will highlight basic molecular and network insights in the autophagy system that are critical in life processes and multiple disease areas, and will examine the prospects for therapeutic targeting of the autophagy system. The meeting will bring together a variety of investigators exploring different facets of autophagy, and their interactions will expand our knowledge of fundamental autophagic mechanisms in different physiological and pathological settings.

Asthma affects people from childhood to old age and causes significant public health problems with great economic consequences. The type 2 inflammation pathway is considered the dominant mechanism of asthma, but clinical trials of recently developed treatments targeting this pathway indicate that large numbers of asthmatics do not benefit from this therapeutic approach. This limited effect of type 2 inhibitors, coupled with recent advances in our understanding of the clinical diversity of asthma, underscores the heterogeneous nature of the underlying disease mechanisms. The aims of this meeting are to focus on the heterogeneous disease biology of asthma and to explore how treatments can advance from a one-size-fits-all paradigm to one in which asthmatics receive personalized treatments based on biomarkers that classify their specific disease mechanism. The meeting program will combine cutting-edge presentations on the molecular mechanisms of inflammation and remodeling in asthma with others that address the complexity of clinical phenotypes of asthma, and will include updates on the application of new technologies to develop biomarkers to guide personalized treatment. Anticipated outcomes include greater understanding of mechanisms of disease other than type 2 inflammation and appreciation of the need to forge stronger collaborations between clinical and basic scientists to advance the goal of personalized asthma treatment. The overarching goal of the meeting is to bring together world-class researchers with expertise in immunology, cell biology and asthma disease biology to permit exchange of ideas and facilitate development of new scientific directions and therapeutic approaches for asthma.

A clear understanding of the mechanisms of host immunity to virus infection will guide efforts to design better treatments for some of the most devastating diseases of humanity. The fields of virology and immunology often convene their own specialized meetings. However, each of these fields has matured to the point where the most transformative advances are likely to arise from cross-disciplinary insights, rather than from continued progress in an isolated area. This meeting will foster new interactions and generate new ideas by assembling international experts in the areas of evolutionary perspectives on the host-virus relationship, innate immune detection and control of endogenous and exogenous viruses, memory cell control of viruses at mucosal surfaces, immunological consequences of prior viral exposure, and new approaches to vaccines and therapeutics. This comprehensive and inclusive approach should spur new thinking. For example, a solution to the medically important issue of chronic viral infection and immune exhaustion may emerge from a better understanding of the history of these viruses and how they have shaped the immune response and vice versa over evolutionary time. Similarly, a rational approach to vaccine design must be rooted in the innate immune mechanisms by which cells detect and respond to virus infection. This Keystone Symposia meeting offers a unique opportunity to bring about the next advances in viral immunity through cross-disciplinary interactions.

The evidence base and research agenda for malaria elimination and eradication are fast-evolving. Of foremost concern is the threat of resistance of the mosquito to pyrethroid insecticides and emerging data showing multi-drug resistance in Southeast Asia. An evolving area, with much to be understood, is the epidemiology of sub-microscopic infections and how they fuel onward transmission. There is also a lot of progress; for example, in the development pipeline there are candidates for a single-dose cure that can also provide protection and block transmission, and there are novel classes of insecticides. Finally, in implementation science, there is plenty to examine, from mass drug administration of anti-malarials to re-purposing ivermectin to reduce residual transmission. This Keystone Symposia meeting examines the scientific progress being made toward the goals of eliminating and eradicating malaria, from biological challenges and discoveries, to the development of the next generation of tools, to potentially transformative strategies to eliminate malaria. The ultimate goal of the meeting is to provide a unique and needed space for the malaria community to challenge hypotheses and share emerging data and insights from the different disciplines in a retreat-like setting where scientists can network, think creatively and exchange ideas. For the sustainability needed to meet the challenge of eradicating malaria, fostering these fruitful exchanges and lasting scientific partnerships is absolutely critical.

A significant challenge in clinical medicine, particularly in cancer and neurodegenerative disease, is the absence of pharmaceuticals that directly target the pathology. In cancer, well over 90% of mutations found in patient tumors are deemed ‘non-actionable’. Many proteins with ‘non-actionable’ mutations are investigated by scientists who study RNA biology, in particular abnormalities in post-transcriptional RNA processing. Biophysicists and engineers have made great strides in understanding biological phase transitions and the role of intrinsically disordered protein (IDP) domains, which have been implicated in both diseases and often described as ‘non-actionable’. This conference seeks to bring together disease-focused medical scientists, RNA biologists, and biophysicists in a truly multidisciplinary environment.

The ability to analyze large numbers of lipids from different categories (e.g., fatty acyls, glycerolipids, sphingolipids and sterols) has greatly advanced our understanding of their biochemical functions and roles in disease, but has also created challenges of scale. This Keystone Symposia meeting will cover areas of lipid research that have been particularly impacted by these technologies. It will specifically address the challenge of managing the large data sets generated by lipidomic technologies and conceptually studying them given how interconnected and cross-regulated lipid metabolic pathways are. Bringing together an outstanding group of scientists with expertise in the lipid categories as well as in lipidomic approaches, it will summarize recent findings for the major lipid categories from state-of-the-art technologies and lay the foundation for a broader understanding of new methods and the technical capabilities and limitations of these methods, as well as where better approaches are needed.

Fibrosis is a common response of many organs to chronic injury and inflammation. This leads to destruction of organ architecture with loss of function. Until recently, there have been no therapies to slow the progression of fibrosis and to maintain normal organ function. Recent advances have provided new insights into the pathogenesis of fibrosis, new drug targets, and new drugs in clinical studies. This conference will bring together basic biologists and translational and clinical researchers to discuss core mechanisms underlying inflammation and fibrosis, to compare and contrast fibrotic diseases, preclinical models, the potential for regeneration and regression of fibrosis, and potential therapies. The interaction between the host genetics, the environment and the microbiome in the pathogenesis of fibrotic diseases will be addressed. Also, non-invasive methods to assess fibrosis including advanced imaging and intermediate biomarkers will be discussed. This conference will be a unique opportunity for investigators who are focused on organ-specific diseases – i.e., medical subspecialists – to participate in cross-disciplinary, multi-organ discussions in order to gain new insights into their research.
