Cell Bio 2022 offers eight different tracks—seven scientific and one covering educational, professional development, diversity, and inclusion.

Toxic Metabolites in the Biology of Ageing and Cancer

This Workshop will focus on the biology of toxic metabolites in the ageing process, stem cells and cancer. Specific emphasis will be placed on how metabolites that carry carbonyls (such as aldehydes and ketones) damage stem and cancer cells. A particular area will focus on endogenous aldehydes (such as acteladehyde, formaldehyde and methyl-gyoxal), the enzymes that remove them [aldehyde dehydrogenases (Aldh) and functionally related glutathione-based enzymes such as Adh5 and Glo-1/2]. Further, we will consider how human genetic variation can contribute to metabolite sensitivity, and how mutations in cancer genomes in toxic metabolism pathways can be exploited for therapy.

Rapid advancement in the field of NMR spectroscopy in recent years has allowed for the development of novel methods for characterizing biomolecular structure and dynamics. The unique ability of NMR to probe biomolecules under native conditions and near physiological state has enabled researchers to address new problems in biology. This India | EMBO Lecture Course will focus on emerging developments in NMR that will spur further innovation in the areas of protein dynamics, membrane proteins, intrinsically disordered proteins, phase separation, paramagnetic proteins, and large protein complexes. We aim to bring together researchers with diverse interests to realize the full potential of the latest advances in solution and solid-state biomolecular NMR spectroscopy, and to engage early career researchers and leaders in the field in discussions about remaining challenges and the new methods and techniques being developed to address these problems. Bringing together scientists with expertise in different dimensions of NMR will enable facile exchange of ideas, spur collaborative research, and drive future breakthroughs.

Recent technological advances have enabled the combination of neuroimaging and electrophysiological methods with NIBS. EMBO Workshop — The 20S proteasome degradation pathway

In the last decade accumulating evidence have demonstrated that the 20S proteasome can function in a free, uncapped form, leading to the processing and degradation of partially unfolded proteins. Various functional roles have been associated with this activity, including neuronal communication, antigenic peptide production, regulation of protein function through post-translational processing and extracellular activities. However, up until now a community effort to advance this field of research or a platform for exchanging knowledge and ideas was lacking. Therefore this meeting will be specifically dedicated to the independent activity and regulation of the 20S proteasome. We believe that such an event will lead to a supportive and collaborative community and advancement of the research. During the meeting, there will be social events and round-table discussions generating a fruitful platform for establishing international ties and novel collaborations.

Gordon Research Conference — Carotenoids

The 2023 Carotenoid Gordon Research Conference (GRC) is a multidisciplinary meeting that addresses the biology of carotenoids and their derivatives apocarotenoids, in microorganisms, plants and mammals, including humans. Carotenoids are synthesized by all photosynthetic organisms and some non-photosynthetic microbes. Besides their vital role in photoprotection and light-harvesting, carotenoids are precursors of established and novel apocarotenoid hormones and regulatory metabolites involved in many aspects of plant's life, including development and intra-specific communications. Unable to synthesize carotenoids, mammals, including humans, obtain these pigments through the diet and convert them into retinoids (vitamin A and its derivatives). Indeed, biofortification of staple crops with carotenoids remains a major strategy to prevent the overwhelming public health issue of vitamin A deficiency (VAD). In addition, carotenoids are used by mammals as antioxidants and for pigmentation (as in macular pigment of the human fovea). Overall, dietary carotenoids and apocarotenoids provide a plethora of health benefits to humans, modulating vision, cognitive functions, embryonic development and growth, as well as preventing onset and development of chronic diseases.

EMBO Workshop — From molecules to organisms: An integrative view of cell biology

This workshop follows the enthusiastic response to the conference organized in 2017. It responds to the need to revisit the structural and functional organization of cells in order incorporate and address two aspects:

1. New emerging concepts in membrane traffic: The study of membrane compartments has traditionally focused on single organelles or pathways. Yet, new insights into inter- organelle communication, membrane-less compartments, organelle-dependent regulation of metabolism, physical principles such as self-organization and mechanics in membrane structure and function, prompt us to pursue a more integrated view of sub-cellular organization.

2. Integrate membrane traffic and membrane biology to a higher level of organization, such as tissues and organisms. Cell biology has to go beyond cells in culture and contribute to the analysis of multi-cellular organisms, during development and in the adult. In this workshop, we aim to gain an integrative and multi-scale perspective of the molecular mechanisms from the sub-cellular to tissue/organism levels. We propose to organize this innovative workshop that can help addressing these challenges. It will therefore focus on new research advances on cytoplasm organization both at the molecular level and subcellular level and bridge it to adjacent fields, such as disease, physics, metabolism and tissue development.

Exposures to environmental and endogenous DNA damaging agents produce thousands to over a million DNA lesions and chemical modifications to the genome per cell every day. Perturbations in the cellular pathways that respond to DNA damage are linked to myriad human diseases across nearly every cell and organ type. Conversely, dysfunction in DNA damage responses presents new vulnerabilities that are being leveraged to selectively target tumors in a more efficacious and less toxic manner. The India-EMBO Lecture course on “Modeling development and disease with human tissue organoids" will bring together researchers working on diverse systems using interdisciplinary approaches to develop organoids to address key questions of human development and disease. Speakers will introduce this frontier area of research, and associated technological and scientific advances, to student and postdoc trainees via lectures and training sessions. This will be the first meeting in this research area to be organized in India and will be of tremendous value in terms of capacity-building and training. Major themes will include neural (neocortex, hippocampus, hypothalamus, retina) and non-neural (kidney, pancreas, skeletal muscle) organoids with a focus on technological advances in bioengineering, imaging, and transcriptomic and epigenomic analyses, which have led to unparalleled new insights into organ function. A key topic of discussion will be the use of organoids to model disease conditions, such as cancer and neurological diseases, which has the potential to transform personalized medicine.

This will be the 4th Conference on Bacterial Cell Biology. In the last two decades there has been an explosion of interest in fundamental aspects of bacterial structure and function, based on advances in fluorescence and cryo-EM imaging. Quantitative and automated methods have brought about radical changes in the breadth and depth of questions that could be explored, while genome sequencing and genetic methods such as CRISPR-Cas9 and Tn-Seq have greatly expanded the range of organisms that can be studied incisively, enabling comparative studies of processes such as growth, morphogenesis and division, across diverse bacterial phyla. Many of these bacterial cell biology systems are being successfully reconstituted using purified components, leading to the discovery of molecular mechanisms underpinning cellular functions. The conference will bring together presentations illustrating the application of advanced cell and molecular biology methods to a wide range of basic functions in bacterial cells. Advances in understanding of the fundamentals of bacterial structure and function also create opportunities for the development of novel approaches to kill or modulate the growth and behaviour of bacterial cells.

The well-studied process of mRNA translation is fundamental to all biology, and the relevance of this process to disease is emerging rapidly. The 2023 Gordon Research Conference (GRC) on Translational Machinery in Health and Disease will provide a unique forum for both new and established investigators to discuss their latest findings on the relationship of protein synthesis to disease. This interdisciplinary conference will cover connections between translation and a broad range of diseases including cancer, neurological, metabolic, inflammatory, infectious, and mitochondrial disorders. Scientific sessions will highlight recent discoveries on the genetic and molecular disturbances of translation that underlie pathophysiology. Other talks will focus on new technologies to study translation, translation during stress, in pathogens, and in the microbiome. A major goal of this conference will be to foster interactions among scientists at all levels to stimulate data exchange and collaborations across disciplines. Ample time will be provided for formal discussions during the scientific sessions and time will also be set aside for informal discussions. Poster presentations as well as the selection of speakers from submitted abstracts will ensure participation from investigators at all stages of their career. In addition, this GRC will be preceded by a Gordon Research Seminar (GRS) on Translational Machinery in Health and Disease which will be open only to students and postdoctoral fellows. The GRS will complement the GRC and provide a forum for trainees from around the world to broaden their peer network and to explore new directions in translation research.

Developmental neuronal remodelling is essential for establishing functional nervous systems across species. Remodelling involves specific elimination of existing connections, typically followed by strengthening of surviving synapses or even axon regrowth to develop new, adult-specific connections. While altered remodeling has long been suspected of contributing to neuropsychiatric conditions, such as autism, schizophrenia, and ADHD, studies only recently established firm molecular similarities between such disorders and developmental remodeling. Moreover, the mechanisms involved in developmental remodelling appear to be reiterated in neurodegeneration and during regeneration. Despite these significant advances in the field, many important questions remain open and will be discussed at this meeting.

Life, as we know it today, is characterized by a set of biophysical properties that include the ability to grow, divide and replicate, and evolve in complexity via selection and distinction. From a chemical perspective, such processes are realized by chemical reactions and assembly processes that turnover reactants, and this turnover is driven by the supply of mass and energy. From a theoretical perspective, this supply maintains the system away from thermodynamic equilibrium leading to continuous fluxes of energy and mass, even in the stationary state of the system. To engineer living systems or specific life-like processes, unified approaches are necessary that incorporate concepts from biophysics, system chemistry, and theoretical physics. This workshop aims to bring together scientists from these three fields to discuss the current challenges to engineering life-like processes. The workshop will provide a collaborative environment that includes plenary talks on the question “What is life?” and the related challenges to engineering it. Moreover, there will be sessions with interactive short talks and brainstorming sessions with plenty of time to freely gather and develop novel approaches to how to engineer life.

Hetero-bifunctional protein degraders like proteolysis targeting chimeras (PROTACs) have found their way into the clinic by utilizing the body’s natural disposal system to tag and degrade unwanted proteins implicated in various diseases. These degraders are large molecules consisting of a E3 binding ligand and a ligand binding the target protein, coupled together by a linker. On the other hand, monovalent degraders or molecular glues consist of a single degrader molecule that attaches to both the E3 ligase and target protein and tend to be smaller and more drug-like. These glues have been discovered serendipitously in the past, however recently, there are efforts being made to try and design them more rationally. Cambridge Healthtech Institute’s inaugural virtual conference discusses some of these innovative design strategies and screening methods to discover and optimize molecular glues for therapeutic purposes. The conference features live talks given by experts and includes online discussions for active networking and idea-generation.

Regulation of translation is key in the control of gene expression, playing a critical role in many biological and pathophysiological settings. In the last decade, methodological advances have provided comprehensive insights into cellular transcriptomes and proteomes, however, the systems-wide analysis of translation remained an unsolved experimental challenge. The advent of ‘ribosome-profiling’ provided the methodological basis for comprehensive transcriptome-wide analyses of translation at sub-codon resolution by the precise and high-throughput determination of ribosomal positions on mRNA. Its application in a variety of organisms yielded novel insights into protein synthesis and its dynamic regulation, significantly advancing our understanding of the post-transcriptional regulation of gene expression.

Ferroptosis is a regulated necrotic cell death modality which is now recognized as an evolutionary vulnerability caused by the incorporation of oxidation-prone polyunsaturated fatty acids (PUFAs) into cellular membranes. This metabolic cell death process provides the framework to understand the basic biological phenomena of how cells restrain, repair and adapt to phospholipid damage through changes in cellular plasticity. A close interplay between cellular metabolism and ferroptosis was established with phospholipid repair linked to numerous metabolic networks including amino acids, glutathione biosynthesis, NAD(P)H and PUFA metabolism, iron handling and the mevalonate pathway. Notably, this complex network has been found to be dysfunctional in different pathological contexts. This EMBO workshop will bring together scientists from different disciplines interested in the molecular mechanisms linked to ferroptosis, such as lipid signalling, redox chemistry, drug discovery, tumour biology, degenrative diseases, medicine and inflammation, and related experts in cell metabolism. This highly interdisciplinary meeting will promote the exchange of ideas and stimulate a critical assessment of the state-of-the-art between students, postdocs and leading researchers from different fields worldwide.

The Hedgehog morphogen pathway is a key signalling pathway in development and stem cell regulation. Hedgehog deregulation leads to developmental and neurological diseases and is a crucial driver of several cancers. The goal of this conference is to bring together experts working on the mechanism of action of Hedgehog signalling in Developmental Biology, Disease, and Signal Transduction. The meeting will focus on the recent structural, biochemical, and cell-biological studies that shed new light on the mechanisms by which the Hedgehog signal is received on target cells and the unexpected role of cholesterol as a second messenger. Moreover, the recent advances in understanding the molecular mechanisms underlying the Hedgehog morphogen biosynthesis, its release from cells into extracellular space, and the mechanisms involved in ligand reception will be discussed. While alterations to DNA underlie genome evolution and antibody diversification, genome instability drives many disorders, including cancer, developmental abnormalities, and neurodegeneration. DNA damage and other genome-threatening replication blocks are generally overcome by a number of DNA repair pathways. The choice of DNA repair pathways, specifically the reliance on error-prone vs. error-free is both intriguing and confounding. Moreover, the deregulation of repair pathway choice can trigger pathological processes. In this conference, we will discuss the delicate equilibrium of genome maintenance pathways, addressing how natural processes such as replication and transcription can threaten genome stability, how the genome can be altered in cancer, signatures of genome instability in human disease, and how cells control the inherent plasticity of their replication and repair machinery. We aim to bring together scientists from diverse fields who are interested in understanding the many processes impacting DNA metabolism and the molecular and biophysical principles that maintain the balance between genome stability and plasticity.

Cell polarization is an essential first step for many biological processes ranging from cell motility to tissue and organism morphogenesis. At the same time, cell polarization requires a precise control of their internal organization, which is the result of a complex dynamic interplay between the cytoskeleton, molecular motors, signaling molecules, and membranes. Recent technological advances have helped obtaining valuable new information on the different temporal and spatial scales that govern cell polarization. We have obtained structures of membrane-localized protein complexes, demonstrated the emergent self-organizing properties of proteins and cellular systems and shed light on how to modulate the shape of membranes and tissues to build entire tissues or organisms. To develop a mechanistic understanding of these processes, we need to integrate our existing knowledge on cell polarization, from the tissue level down to individual molecules, obtained from minimal reconstituted systems to whole animals, from experimental and theoretical approaches. This EMBO Workshop will bring together a highly diverse group of scientists (cell & structural biologists, (bio)chemists, experimental & theoretical physicists) to present the state-of-the-art of their fields, to exchange ideas, and to identify synergies between them to establish new collaborations and open new research fields.

RNA and protein degradation play critical roles during dynamic proteome remodeling in response to developmental and environmental cues. Although RNA and protein degradation pathways have often been investigated in isolation, there is a growing interest in understanding how RNA decay and protein degradation systems interact and cooperate to shape both the RNA and protein landscape within cells and tissues. For example, an explosion of recent research has demonstrated that RNA decay and protein degradation cooperate when ribosome progression is halted to facilitate removal of potentially toxic mRNA and/or protein species. Further, regulating the abundance of either RNA or cognate RNA binding proteins can alter the behavior of highly dynamic macromolecular assembles that can form liquid-like condensates and have been linked to various human pathologies. The purpose of this EMBO Workshop is to bring together experts in the RNA decay and protein degradation fields to report on our current understanding of RNA and protein degradation pathway intersections, discover common principles, and share technical expertise on best practices to interrogate RNA or protein turnover.

After recent advances in the field of molecular biology and genetics accompanied by the breathtaking progress in multi-omic strategies, have led to the development of Systems Biology in the last 5 years. Holistic approaches enable a refined approach to study the dynamic interactions between the components of the nucleus, the cell and its environment, ultimately leading to understanding the biological networks and the properties of biological systems. Recent advances, such as reaching the unprecedented resolution of individual cells combined with the use of new systems biology approaches, allow to address long standing questions, especially in the fields of disease and development. Thus, this EMBO Workshop will bring together world-class experts and young scientists in the fields of Systems Biology, Chromatin and Epigenetics for four full days to discuss their exciting recent findings as well as the future directions of these fields. The topics of the EMBO Workshop are sufficiently covered by the outstanding speakers. This EMBO Workshop has a unique focus and will allow all participants to receive a coherent, updated and original view of how the interplay of Systems Biology, Chromatin and Epigenetics will advance our understanding of development and disease, important areas that hold the keys for future therapeutic approaches.

The discovery of myriad noncoding RNAs (ncRNAs) over the past decade has revealed a universe of new and exciting therapeutic opportunities. Many thousands of microRNAs, piRNAs, circular RNAs, long noncoding RNAs, and others are expressed across healthy and diseased human tissues and play key functional roles in pathogenic states and processes. Their regulatory roles, cell-type-specificity, and vast numbers confer on ncRNAs unique advantages as therapeutic targets. Rapid advances in clinical transcriptomics, oligonucleotide therapeutics, and RNA-directed small molecules, open the possibility for rapid, economical, and personalised ncRNA-directed therapies. Reaching this goal will require overcoming key challenges, including low evolutionary conservation in model species, poorly-characterised molecular mechanisms, finding hits amongst the vast ncRNA transcriptome, and effectively drugging targets in vivo. This EMBO Workshop will bring together colleagues from both academic and commercial worlds to discuss their latest progress in these issues. Sessions will cover the full span from basic disease biology to therapeutic development. Confirmed speakers represent leaders from around the globe, and numerous slots have been set aside for oral presentations selected from submitted abstracts.

Advanced reproductive technologies, such as intracytoplasmic sperm injection, have revolutionized reproductive medicine by enabling effective fertilization without the need to understand the cause of infertility. However, declining male fertility persists in many countries, and growing evidence suggested that the reproductive health of fathers affects their children’s health, calling for continuing investigation of the underlying mechanisms. The main themes of this EMBO Workshop are the male reproductive system, testis development, and sperm formation in mammals, including humans and animal models. Reflecting the multifaceted nature of male reproduction and discoveries made by increasingly advanced experimental techniques, the three broad topics further branch into sub-topics including genetics, epigenetics, immunity, lifestyle influences, toxicity, immunology, and endocrinology. Clinically relevant topics—semen function, sperm physiology, reproductive technologies, and fertility preservation—are also incorporated. This EMBO Workshop aims to bridge the knowledge gap in male reproduction, facilitate networking among research groups with different expertise, and promote the translation of fundamental research in male reproduction into diagnoses and therapies.

Meiosis is a defining feature of sexual reproduction. This specialized cell division program comprises a single round of DNA replication followed by two successive cell divisions, thereby reducing chromosome number by half. This reduction accommodates the later combination of two haploid gametes during fertilization to reconstitute a full diploid genome. The unique events of meiosis, including pairwise alignment of parental chromosomes, inter-homolog recombination, kinetochore attachment, cell cycle adaptations and error correction mechanisms, are intensely studied processes in numerous model organisms (fungi, protists, plants, and animals). The Meiosis field is driven by fascination in how evolution has implemented and perfected the meiotic program, and also by its significance to both plant breeding and human health. As a special topic for the 2021 EMBO Workshop, we have chosen homolog pairing. The recognition and physical pairing of homologous chromosomes is an essential feature of meiosis, and also occurs in other contexts. It is therefore timely to bring together the fields of meiotic and post-meiotic chromosome pairing to potentially uncover common principles and mechanisms.

The ubiquitin-proteasome, lysosome and autophagy systems are all well-controlled, selective degradation pathways that are key cellular regulators in cancer, CNS, and other diseases. A new generation of hetero bi-functional chimeras and small molecule monovalent degraders are being developed to hijack these systems for targeted protein degradation or stabilization. These molecules that include Proteolysis-targeting chimeras (PROTACs) and molecular glues are being used to seek out previously “undruggable” targets for therapeutic intervention. However, challenges do exist in terms of specificity, stability, biodistribution and penetration of these degrader molecules. This conference on Next-Generation Degraders & Glues brings together experts in the field to discuss these new therapeutic modalities, as well as issues underlying the use of targeted degradation as a new therapeutic approach.

Viruses are major human pathogens that cause a wide range of infectious diseases, representing a constantly recurring challenge to the health of individuals and the functioning of human society. Only a profound understanding of viral biology will provide the basis for developing and improving antiviral therapies. While RNA turnover has long been studied in the context of cellular gene regulation and quality control mechanisms, the interplay between RNA turnover and viral life cycles is only now emerging as a key component of virus-host interactions. For example, Herpes viruses, Poxvirus, Influenza virus and SARS-Coronavirus encode ribonucleases that cleave host mRNA and thereby support virus reproduction. Flaviviruses such as Dengue virus, West Nile virus and Hepatitis C virus encode a non-coding RNA that blocks Xrn1, a major cellular exoribonuclease, as a means to escape from an antiviral activity. The endonuclease RNase L is another component of the mammalian antiviral defense system, highlighting the importance of ribonucleases in the encounter of viruses with their host cells.

This EMBO Workshop will bring together molecular biologists working on basic mechanisms controlling RNA turnover with virologists exploring RNA turnover in the context of virus-host interactions, and propel the scientific exchange in this emerging area of research.

In recent years nucleic acids have seen wide use in chemical biology investigations as well as in the clinic as therapeutics and diagnostics. The antibody response and the complement system are major effector arms of the immune system. As the portfolio of antibody—and complement-effector function grow, high-end technological solutions are developed, and novel therapeutic applications are proposed. This EMBO Workshop on Antibody & Complement will be a forum for leaders in the field and new investigators coming from academia, industry and research institutes to discuss where the field is going and what challenges lie ahead. The conference will provide the platform for researchers in the recombination and repair fields to present their new unpublished data that sheds novel insights into not only the mechanisms of DSB repair by recombination, but also the consequences of inaccurate repair leading to cancer. It is now clear that many inheritable cancers, including those linked to defects in the BRCA1 or BRCA2 genes, are caused by defects in homologous recombination. Understanding the precise mechanisms of recombinational repair is therefore critical for the development of new drugs that can target these diseases and improve human health.

A long-standing question in the neurobiology of learning is that of timescales. The brain can associate events that are separated by seconds, minutes, hours, days, weeks, or longer. Yet, the known biological mechanisms for inducing changes in the synaptic connections between neurons are driven by neural events that are separated by typically no more than a few tens or hundreds of milliseconds. This conference will convene scientists studying the molecular, cellular, and circuit-level mechanisms used by the brain to bridge the multiple timescales across which events play out in the experience of an individual. The goal will be to assemble an up-to-date understanding of the set of biological mechanisms available in the brain to support associative learning across different timescales. The conference's emphasis on biological mechanisms will complement and support the main program’s more computational focus, and encourage an integrated analysis of statistical learning spanning molecular, cellular, circuit, and behavioral-level phenomena.

Mitochondria act at the interface of numerous processes critical for cellular function and organismal health. Long recognized as the powerhouses of the cell, mitochondria are emerging as hubs for cellular metabolism, redox balance, signalling, gene expression, detoxification, and ion homeostasis. Mitochondrial maintenance is critical for proper development and function of cells and tissues, and mitochondrial dysfunction underlies numerous disease processes ranging from severe childhood mitochondrial disorders, to heart disease, neurodegenerative disorders, cancer, and aging itself. This conference will bring together scientists and clinicians with expertise in a breadth of mitochondrial biology to explore topics at the interface of basic and translational biology. Mitochondrial basic mechanisms of organelle regulation, mitochondrial mechanisms of disease, cutting edge technologies, and interventional strategies will be emphasized in interactive scientific sessions.

The germline provides an eternal cellular link between generations and is the source of genetic diversity and evolution of animal populations. In addition, perturbations in the epigenome or genome of a parental germline that arise during development could be underlying causes of many disease states in offspring. In males, sperm are the conduit for transmission of genetic and epigenetic information to the next generation and are formed by the complex process of spermatogenesis, which involves undifferentiated stem cell, differentiating, meiotic, and morphogenesis phases. This workshop will explore recent advances in understanding the biology of these different phases in a variety of animal models as well as how epigenetic and genetic abnormalities may underlie transmission of diseases and fertility disorder in humans. Invited talks and poster sessions that focus on cutting edge findings along with vibrant discussions and debates about controversies in the field will provide a rich intellectual atmosphere that is sure to have an important impact on how the biology of spermatogenesis is investigated for years to come.

The maintenance of genome integrity is critical for the suppression of several pathological disorders in humans, including cancer, infertility and neurodegeneration. Moreover, the accumulation of unrepaired errors in DNA is commonly cited as a likely cause of tissue and organismal ageing. Destabilization of the genome can occur as a result of several cell intrinsic or extrinsic factors, including errors arising during DNA replication or chromosome segregation, as well as exposure of cells to agents that induce DNA damage. In this conference, we aim to bring together scientists studying DNA replication, DNA repair and chromosome segregation with those interested in how chromosomal instability can influence human pathology. Moreover, we aim to show how high throughput and high content screening methods can be used as a discovery tool both for basic science applications and to identify potential therapeutic modalities.
