During autophagy, cytoplasmic components ranging from individual proteins to organelles and pathogens are delivered to the lysosome or vacuole for degradation and reuse. The discovery of a set of conserved ATG proteins involved in autophagy in yeast, recognized by a Nobel prize in 2016, established the core machinery. The complexity of autophagy processes is dramatically increased in higher eukaryotes. Autophagy is essential for the homeostatic balance of cells and organisms and its deregulation has been associated with a plethora of human diseases. There is an unmet need to deepen our understanding of its roles and molecular mechanisms, its regulation by various signaling pathways during normal and stress conditions, and its targetability for therapeutic intervention strategies. The goal of this meeting is to expand and integrate new and unpublished data on the genetics, proteomics, cell biology, biophysics and biochemistry of autophagy processes with previously under represented model systems, disease relevant topics and emerging technologies. The meeting will foster interactions between early-stage researchers, junior group leaders and established investigators and thus facilitate the enrollment and education of the next-generation scientists to tackle challenges implicated in the translation of basic research findings towards disease models diverse in scale and species.

High-throughput sequencing results in recent decades have revealed that transcription of the eukaryotic genome goes beyond coding regions and uncovered an overwhelming amount of novel functional RNAs. The list of these RNAs is constantly increasing and includes lncRNAs, circular RNAs and enhancer RNAs, among others, with crucial roles in genome organization, transcriptional and posttranscriptional gene regulation. The basic mechanisms of action of these RNAs are only beginning to emerge and are most likely exhibited due to specific RNA secondary and tertiary structure, as previously found for other RNAs, e.g. ribosomal RNAs and tRNAs. Nevertheless, the connection between RNA structural biology and RNA function is still in its infancy. To deeply understand how molecules work, detailed knowledge of their structures is necessary.

The most important factor that limits the success of anti-cancer therapy is the emergence of drug resistance. At a time where we seem to have reached a limit in terms of genetic mutation driven-treatment efficacy in multiple cancer types, i.e. with a best overall survival around 30-36 months in melanoma but still more than one third of deaths after two years, there is an urgent international need for innovative strategies to combat the resistance. The milestone discovery of drug-tolerant cancer persistent cells suggests that drug resistance might derive from genetically similar clones. Indeed, advances during the past decade show that persistent cancer cells are the discrete and usually undetected cells that survive cancer drug treatment and constitute a major cause of treatment failure. This workshop will provide a unique opportunity to promote communication among biologists, mathematicians and clinicians, which will enable interdisciplinary discussions and collaborations for this fasting emerging domain. The sessions will start with understanding molecular mechanisms of cancer persistence, and will continue with discovering therapeutic targets of persistent cells, deciphering their heterogeneity at single cell level, learning from the microbial persister world, and modelling of the dynamics of persistent cells.

The kinetochore is a dynamic proteinaceous structure that forms at centromeric chromatin. It connects spindle microtubules to chromosomes in order to ensure the equal segregation of chromosomes during mitosis and meiosis. Over the years, the Dynamic Kinetochore meeting has become the major venue for discussing the molecular details of kinetochore formation, structure and control and their relevance to cancer and other human diseases. The meeting encompasses the latest unpublished and cutting-edge research in the field and includes topics that span different disciplines (from biochemistry and biophysics to animal biology), and organisms (from yeast to human). Traditionally, the meeting fosters early career principal investigators, students and postdocs but also features key senior members of the field.

Time is inherent to biological processes. It determines the order of events and the speed at which they take place. This is particularly obvious during embryonic development where the sequence and rate of events ensures that structures develop in the right place, at the right time. Modifying the developmental tempo can affect the final size and composition of tissues, as well as their plasticity and function. Strikingly, comparisons between species reveals that, although the order and underlying molecular mechanisms are often indistinguishable, the pace at which they advance can differ substantially. Hence comparison of biological processes between species is likely to provide insight into time-keeping mechanisms and how they change. Despite the centrality of these questions to embryo development, and evolution, it has remained an understudied topic. This conference will foster dialogue between researchers from different fields to stimulate new ideas and catalyse the study of the genetic, molecular and cellular mechanisms controlling the tempo of embryonic development.

Microfluidic tools have made recent biological and biomedical research leaps possible. At the physics, engineering, chemistry and biology interface, innovation in microfluidic approaches has positively impacted areas as diverse as nucleotide sequencing, functional genomics, single-cell and single-molecule studies, and biomedical diagnostics. Many of these applications – including next-generation sequencing devices – have been revolutionised by miniaturisation, paving the way for global gene analysis and hence transforming biology. Working on the pico- to nanoliter scale not only allows a high degree of parallelisation, but importantly offers superb spatiotemporal control. Small objects such as cells, or even discrete parts thereof, can be exposed to unique conditions, facilitating entirely novel approaches to inquiry in modern biology and chemistry. The EMBL Microfluidics Conference brings together top researchers and emerging research leaders to spark scientific exchange and create community. Topics spanning from fundamental physics & chemistry to device design and nascent biological applications will be presented, which should be of interest to everyone from experts in microfluidic design to users of the next-generation of microfluidic tools, and from academic scholars and trainees to industry colleagues.

Eukaryotic cells maintain their genome as linear chromosomes in the nucleus and smaller, circular chromosomes in their autonomous organelles. Sporadically, they also contain non-chromosomal DNA, generally in circular form, due to recombination of chromosomal fragments, movement of DNA from mitochondria to the nucleus, uptake of extracellular DNA, or the presence of selfish DNA of endogenous or exogenous origin, like transposons and viruses. When it does not harm or kill the cell, this material may contribute to the evolution and structural dynamics of the genome. Reversely, selfish DNAs evolve sophisticated machineries to multiply and propagate independently of, but also frequently via chromosomes. During this meeting we will review recent insights on the complex relationships that chromosomes and extra-chromosomal DNAs entertain with each other, discussing how cells distinguish them, how their relationships contribute to the biology of the cell in health and disease, and affect genome plasticity and evolution. The intricate relationships between chromosomes and extra-chromosomal DNAs are raising fundamental questions in both basic and medical sciences. We will further conceptualize these questions and discuss how to best approach them experimentally and theoretically.

This symposium provides a discussion platform for all scientists interested in the biology of infection. Cell biologists, microbiologists and immunologists will discuss in joint sessions recent progress in our understanding of host-pathogen interactions. Talks will focus on common processes, rather than particular infectious agents or host cell types, and the strategies of all major types of pathogens in their interaction with plants and animals will be covered. A better understanding of infectious diseases will come from the simultaneous analysis of microbial and host variables, which may open new avenues for treatment and prevention strategies.

RNA is one of the building blocks of life and the mechanisms underlying RNA metabolism are critical for all cellular functions. In recent years, exciting discoveries have shown that targeting mRNAs to subcellular compartments and local translation plays a major role in controlling protein function from unicellular organisms to humans. However, the molecular mechanisms regulating mRNA localization and local translation, and their coordination to produce complex cellular behaviours, remain poorly understood. Interest in this field has increased with high-profile discoveries of liquid- or gel-like properties of mRNPs and the relationship of altered RNA granule assembly and localization to neurological and ageing-related disorders. Remarkable technological developments have allowed visualization of individual transcripts and their translation in single cells and in living organisms. This EMBO Workshop will bring together scientists from different fields and with complementary technical expertise to discuss recent discoveries in mRNA localization and local translation and build new collaborations to advance our understanding of these processes.

EMBO Practical Course — Integrating traditional and molecular approaches in ecology of glacial habitats

Glaciers have long been considered almost lifeless environments, but this view does not correspond to reality. In recent years, they have been recognized as ecosystems in their own right, teeming mostly with microbial life, but also hosting larger organisms. Our current knowledge of glacier ecosystems is poor and fragmented. Most studies conducted so far focused on peculiar supraglacial habitats and were restricted to specific taxa. The course is organised in two locations and will last 12 days. The venue for field activities and some lectures is the Rifugio Branca in Santa Caterina Valfurva (SO), Italy Alps. Alpine Guides and expert researchers will guide participant for sample collection in the National Stelvio Park. The venue for wet molecular laboratories, bioinformatics and statistics computer laboratories, practicals and other lectures is the University of Milano Bicocca, Milan, Italy.

Light sheet microscopy is an established imaging technology that enables the recording of large biological specimens with minimal photo-damage. It opens new avenues to study cell biological and developmental processes with unprecedented imaging speed and facilitates systems biology approaches by quantitative imaging of living systems in their entirety. Although light sheet technology is still developing, it has become broadly available through commercial and open-access platforms. In this interdisciplinary course, we are continuing the tradition of three such courses in 2014, 2016 and 2018 at MPI-CBG in Dresden. This new installment of the course at CEITEC Brno will focus on the emerging trends in the field, namely, the combination of light sheet imaging with other imaging modalities in a correlative approach and probing the physical properties of living systems imaged with the light sheet technology. Given the scientific focus of the hosting organisation, we will particularly welcome students bringing applications of light sheet microscopy to imaging plants and viral infections.

We are living in an extremely interesting period in cancer research. Knowledge gained in the last decades is now entering the clinic and new findings are translated into the clinic at a pace never seen before. Most of the successes come from our understanding of basic processes that govern cancer initiation and progression, including signal transduction. Consequently, we see that especially inhibitors to signalling molecules, in particular kinases, are entering the clinic very rapidly. Therefore, the molecular mechanisms by which signals are received by cells, transduced towards the proper targets and integrated into a biological response is amongst the most-studied areas in modern biology. In this EMBO|FEBS Lecture Course we are bringing together a number of true experts in the field from basic and translational oncology. This MBO|FEBS Lecture Course will cover signalling by various pathways (PI3K, WNT, RAS/RAF) but also DNA damage and protein degradation signalling. We will also cover the consequences of pathway deregulation for cellular processes like metabolism and damage repair. Finally, the application or translation of this knowledge into the clinic will be discussed by the speakers.

The concept of “characters” lies at the heart of many central issues in evolutionary biology. Characters distinguish taxa, cause fitness differences, and generally are the fundamental units of comparison for studies of phenotypic evolution. Historically, the concept of characters originated in morphology and comparative anatomy, where it was intimately connected to the issue of homology. The summer school is mainly aimed at early-stage (PhD or postdoc) empirical and theoretical researchers with a general background and/or interest in developmental and/or evolutionary biology. More senior investigators are welcome to apply as well. Exceptions can be made for motivated masters students. Participants of previous Venice Summer Schools in Evo-Devo are expressly encouraged to reapply as this course has an entirely different topic than previous editions. The course will equip participants with the conceptual tools to engage in a productive discussion of the notion of “characters,” and to relate this notion to their own research questions and explanations.

The EMBL Transcription and Chromatin meeting has a long-standing tradition in shaping the field of transcriptional regulation. The meeting brings together leading experts covering all aspects of transcription including cis-regulatory function, long range regulation, 3-dimensional looping, the basal transcriptional machinery, RNA polymerase regulation and function, nucleosome positioning, chromatin modifications, chromatin remodelling and epigenetic inheritance of transcriptional silencing. The meeting contains many talks selected from the abstracts that are interspersed with invited speakers, discussing the latest breakthroughs in transcriptional regulation. The conference is designed to promote interactive discussions at both the talks and poster sessions. Given the excellent line up of speakers and the meeting’s outstanding reputation, this is a ‘must’ attend for anyone interested in cutting edge research in transcription.

Chromatin structure, organization and dynamics underlie essentially every aspect of cell function. Rapid advances in recent years in imaging- and sequencing-based methods and in structural tools, such as Cryo-EM, resulted in breakthroughs in our understanding of the spatial and temporal organization of chromatin and its functions. The aim of this workshop is to bring together experienced scientists, Ph.D. students, and postdoctoral fellows working on the functional organization and dynamics of the nucleus and chromatin in diverse cellular processes. Our goal is to establish an informal and intimate atmosphere (120 participants in total), in which oral and poster presentations by students and postdoctoral fellows, from selected abstracts, will be complemented by lectures delivered by leading researchers in the field. The speakers will present exciting recent work, discuss their perspectives on future developments, and highlight the existing conceptual views and technical problems in the area. The lectures will be organized in 8 sessions and will be complemented by 2 poster sessions.

EMBO Workshop — When predictions meet experiments: The future of structure determination

Figuring out the “protein folding problem”, that is predicting protein structure from the amino acid sequence, has stood as a grand challenge in biology for the past 50 years. Thanks to the efforts of several generations of structural biologists, bioinformatics experts and artificial intelligence (AI), the organisers of the biennial Critical Assessment of protein Structure Prediction (CASP) competition have declared the AlphaFold program as a solution to this grand challenge. This major breakthrough has demonstrated the impact that AI can have on scientific discovery and its potential to dramatically accelerate progress in fundamental fields. At the same time, however, predictions should not be perceived as a way to substitute experimental work. Important challenges are still ahead of us: intrinsically disordered or multidomain proteins, large complexes. The new tools should be used not to stop structural biology but to push its frontiers. The present workshop is thus meant to fill the gap between experimentalists and computational scientists, provide mutual feedback and constitute a forum to promote and facilitate the dialogue on how AI predictions can be integrated in the normal practice of structure determination to increase the capabilities of both experimentalists and computational scientists.

Since the first meeting in 2014, we have witnessed huge advances in the field, driven largely by technological advances such as organoid culture and single cell sequencing, and have seen significant expansion in the range of questions that can be now addressed using human systems. It is therefore more important than ever to provide a forum for researchers to come together and discuss the latest advances, and remaining challenges, in the field.

Nuclear receptors (NRs) constitute the only reliably druggable class of transcription factors, thereby functioning as both key downstream cellular effectors and direct drug targets. Despite numerous therapeutic innovations and treatments, much remains unknown about the biological processes under the control of NR activity. This EMBO 2022 Workshop on a 20/20 vision of the future of NRs and their role in precision medicine, aims to exploit this unique position of the field (i.e. long track record of singular achievements having high impact in diverse fields of research, remarkable clinical relevance and being at the vanguard of technical and methodological advances) to showcase the future of this important field and to highlight recent advances that will form the basis for the future treatments, diagnostic assays and for assessing the latest innovations. The field is at a critical juncture, and whilst this meeting will continue the very successful EMBO NR conference series, the EMBO 2022 workshop will have a different twist on the theme. Unicellulars represent ~70% of extant eukaryotic diversity and are responsible for the production of half of all oxygen on Earth. Major challenges in the field are underpinned by this staggering diversity of this domain of life, much of which remains uncultured. Still, for the vast majority of microbial eukaryotes, be they terrestrial or aquatic, we know nothing about their interactons with other living entities. Therefore, with this series of EMBO workshops, which was so far extremely helpful in propelling comparative genomics and diversity of microbial eukaryotes, we will now pay more attendtion to microbial interactions, as they truly underpin the survival of all life. In order to understand microbial ecosystems, we need to focus on interactions, ranging from ephermeral to permanent, and their evolutionary, physiological, and ecological implications. These topics will be combined with bringing forward the latest developments in the studies of the last eukaryotic common ancestor and non-traditional model organisms and knowledge of their cellular biology – especially as it connects to interactions with the external world. Inspired by the currect pandemics, we will also pay attention to viruses affecting this domain of life.

Reconstructing the Human Past – Using Ancient and Modern Genomics

The study of population genetic variation and the sequencing of ancient DNA represent promising new avenues for investigating human history and our evolutionary past. Population-scale sequencing projects investigating human diversity have provided us with more than a million genome wide datasets that allowed new insights into patterns of human variation and mobility, while others have obtained genome wide data from thousands of ancient human skeletons, allowing the investigation of human evolution in action and providing direct insights into population genetic dynamics in situ. Large-scale genetic variation data sets can now be integrated with genomics data from ancient remains and history records to provide novel insights into human history, cultural evolution and the genetic history of societies. Ancient DNA was furthermore used to reconstruct genomic variation of historic pathogens as well as oral and gut microbiomes in order to provide molecular fossils to study microbial evolution through time. This meeting will involve scientists from genomics, bioinformatics, microbiology, anthropology, archaeology and historians and may initiate future interactions in this exciting and timely new research area that has the potential to change the way we think about our human past and how we might study genetic variation in the future.

Defining the end of transcription units across eukaryotic genomes is critical to understanding how individual genes are regulated. RNA 3' end processing triggers release of transcripts from their chromatin template, leading to production of messenger RNA as well as triggering degradation of excess transcripts. To fully understand this process it is important to consider how RNA 3' end formation is coordinated with gene transcription and other RNA processing mechanisms. Furthermore the mindshift in considering RNA 3’ ends in the context of the whole genome and its transcriptome requires extensive computational biology/bioinformatics to fully appreciate the genome wide biological roles of this process. This EMBO Workshop will bring together many of the top scientists in this field who employ a combination of molecular biology approaches that also harness recently developed bioinformatics, proteomics and computational based cryoEM. Surprisingly even though aspects of this subject area have been under investigation for decades, only now do we appreciate how critical this process is to gene expression in a genomic context. In particular recent studies show that cellular stress through viral infection or cancer induction both cause a breakdown in RNA 3' end regulation. All of these and more topics will be covered in our forthcoming, intense and in person EMBO workshop on RNA 3’ ends at Brasenose College Oxford this September, 2022.

Over the past decades, we have seen growing evidence that environmental influences such as diet, stress, or trauma, can induce changes in individuals that are inherited by their children and grandchildren, independently of changes in the DNA sequence. This type of inheritance is often referred to as epigenetic inheritance, however, the biological mechanisms behind this phenomenon remain poorly understood. The proposed workshop will bring together experts in the field of epigenetics to discuss how the epigenome is inherited on the cellular, organismal, and population level in diverse biological contexts. Moreover, the workshop will explore how different biological systems achieve the correct balance between epigenome inheritance and reprogramming, and what sort of ramifications this has for developmental processes, disease etiology, adaptation, and evolution. Numerous recent findings in this rapidly developing field have been fueled by the advent of genomics technologies, which offer ever-increasing resolution and precision for epigenome quantification. Our meeting will thus provide a unique opportunity to discuss how these recent advances have reshaped the field and what challenges we might expect as we move forward to unravel how epigenetics participates in fundamental biological processes.

This conference series has become known for exploring yeasts as models to study questions of general relevance to evolution and ecology. Experts will speak on a range of topics, including evolution theory, ecology, genetics, cell biology, genomics, systems biology, metabolic modelling and engineering. The focus of this conference will be on comparative and experimental strategies to explore evolutionary questions, from field studies to computer simulations to comparative genomics. Central evolutionary questions will be discussed, using yeast and a range of other species whose biology is well understood and to determine the degree to which that biology is conserved in more distantly related species. The aim is to move from correlative studies based on comparative work to more causal studies based on broad experimental support. This problem is particularly pressing in the age of cheap and easy sequencing.

Redox reactions are central to all life. It is therefore unsurprising that organisms have evolved numerous strategies to monitor and respond to different cellular redox species. Central to these processes are redox-regulated proteins, which respond to specific redox species via oxidative post-translational modification of defined cysteine residues (thiol oxidation). Protein thiol oxidation is increasingly recognised as an important regulator of many physiological processes whilst aberrant regulation may play a key role in many different pathologies. This EMBO Workshop will bring together researchers employing a broad range of methodologies and model organisms to understand mechanisms and targets of protein thiol oxidation as well the relevance of thiol oxidation for organismal physiology and pathology.

Before the genetic information stored in DNA can be used to direct cell growth and metabolism, it has to be transferred into RNA. Messenger RNAs (mRNAs) that code for proteins and noncoding RNAs are key components in the transmission of genetic information in all life forms – from viruses to complex mammalian organisms. Exciting recent findings now reveal a new layer of information added to RNAs in the form of chemical marks (the epitranscriptome) that play a critical role in gene expression control. Following the great success of the virtual 2020 edition of this long-standing symposium, we are looking forward to returning to an in-person meeting for 2022, bringing together leaders in the RNA field, post-docs and students, with the aim of disseminating and discussing the most recent results. The program will include, among others, sessions on transcription, RNA processing and modification, mRNA export and localisation, mRNA surveillance and decay, translation and the control of mRNA expression by microRNAs.

Metagenomics studies are rapidly uncovering the compositional richness of microbial communities in diverse habitats ranging from the oceans to the human gut. While their fundamental role in our health and environment is undeniable, there is an urgent need for unravelling molecular mechanisms underlying the dynamics of these communities. This requires a combined experimental and computational approach. Recent studies (including those from the organisers’ and speakers’ labs) have underlined the success of such integrative approaches. For example, emergence of cell subpopulations (Varahan et al. eLife 2019, 2020), and long-term stability (Blasche, Kim et al. Nat Microbiol. 2021) through metabolic cross-feeding. As researchers world-wide map species dynamics in the ecosystems of their interest, this course will provide the necessary overview and tools for moving from cataloguing to identifying key interaction agents, especially metabolites. This will include integrating species and metabolite dynamics data, as well as leveraging the power of genome-scale metabolic models to predict community interactions.

This EMBO Workshop will include new discoveries that surround the ER's historical and important role in regulating the translation, translocation and folding of membrane and secreted proteins of the secretory pathway. It will further include new insights into the signaling pathways that allow the ER to correct for cell stress, the unfolded protein response, and maintaining lipid homeostasis. Recent years have also revealed emerging and unexpected new functions for this old organelle in regulating the biogenesis of other organelles at membrane contact sites (MCSs). These MCSs have generated much excitement in cell biology because they are hybrid locations where the elaborate ER network can communicate with other organelles through calcium signaling, lipid metabolism, and the recruitment of machineries that modify membrane shape, trafficking and division. They reveal that not every membrane bound component traffics through vesicles, rather many move directly at positions where the ER is tethered and apposed to the plasma membrane and other organelles to rapidly communicate signals across the cell that can maintain homeostasis. Disruption of ER homeostasis and ER structure is a hallmark of various diseases. Thus, understanding basic mechanisms controlling ER functions may provide insight into a broad range of pathologies.

Cognitive deficits in neurodevelopmental disorders (NDDs) are a significant health burden worldwide. Mechanistic understanding of developmental defects of neural circuitry from system to molecular level is limited. The elucidation of genetic or epigenetic factors modulating the establishment of functional neural circuits during critical periods and their impact on cognitive function has shed significant insights into the causal relationship of behavioural deficits associated with NDD. The seamless integration of molecular knowledge with systems- and cognitive- level analyses is the 'need of the hour' to associate mechanisms of circuit-specific developmental deficits with cognitive outcome linked to NDD. This meeting will bring together the leading experts who are working on various aspects of the NDD to discuss recent advancement in the field. This EMBO Workshop will create a platform for young neuroscientists including postdoctoral fellows and graduate students who wish to pursue their research in this emerging field. This EMBO Workshop will particularly enable postdoctoral fellows and graduate students to present their research in front of leading experts and learn about challenges in the field through an informal, yet an intense discussion with invited speakers.

There has been an increasing awareness in the scientific community regarding the inability of animal models to often capture the complexities of human biology. In the last decade, there have been significant advances in the field of model systems that are based on human biology, including microphysiological systems (MPS). These systems aim to recapitulate the physiology and functions of a single human organ or a network of organs at a smaller scale, such as organoids or organs-on-chip. MPS have often been proposed as the next frontier of human-relevant research in drug discovery and testing. The lecture course will give an overview of the current research of how these model systems are being used to understand human disease and development. It will also cover design considerations, current challenges, setting standards and roadmap for broader adoption and use of these systems. In addition, the course would have networking sessions for early career researchers and students, along with discussions on collaborations, international grants, and entrepreneurship.

EMBO Practical Course — Volume electron microscopy by automated serial SEM

Developments in scanning electron microscopes (SEM) in recent years have led to ground-breaking opportunities in volume imaging of cells and tissues. The aim of this EMBO Practical Course is to provide extensive theoretical and practical knowledge on the three main techniques for automated serial imaging based on SEM (ASI-SEM): microtome-based serial block-face SEM, focused ion beam SEM and array tomography. At the heart of the course, participants will be able to assess the potential of each technique to understand which fits best with their own scientific project. The participants will acquire the theoretical background to understand the different techniques as well as how to optimize the imaging conditions. Data processing, analysis and management, key considerations in all ASI-SEM techniques will also be covered. This EMBO Practical Course has a significant hands-on component where students participate in experimentation on i) sample preparation adapted to each ASI-SEM technique ii) operation of the hardware (SEMs, automated microtomes) and iii) image processing software (stack registration, segmentation, tracing).

This EMBO practical course will give a practical introduction to integrative structural biology. Through lectures and hands-on practicals, the participants will learn about the fundamentals of X-ray crystallography, cryo-EM, small-angle X-ray scattering, NMR, and structural mass spectrometry. The invited tutors will show how to analyze and interpret the experimental data, and how to use the data for building structural models across different resolution scales. The course will also explore how experimental and computational methods aiming at resolving dynamics can be integrated to characterize flexibility in macromolecular structures. Finally, we will discuss the future of integrative structural biology approaches in light of the most recent advances in experimental and computational methods.

The Neurovascular Interface

Neurovascular biology is a young, emerging field that aims to unravel the molecular communication between the nervous system and the vasculature. Both the nervous system and the vascular system share striking conceptual similarities in the assembly of cellular networks through guided growth and branching processes. Vascular and neural cells work together to form a number of functional tissue compartments. Endothelial cells, certain mural support cells (i.e., pericytes), astrocytes and neurons assemble the neurovascular unit, which establishes the blood-brain and blood-retina barriers. In fact, many of the most debilitating neurodegenerative diseases and cognitive decline involve disruption of the neurovascular unit or compromised vascular function in the nervous system. It has become increasingly clear that the endothelium and mural support cells function go beyond nutrient supply and are instructive and structurally required to allow nervous system function. In this symposium, we bring together researchers from vascular biology and neurobiology to foster interdisciplinary exchange and new emerging concepts on how vascular and nervous systems crosstalk to maintain organ homeostasis and function.

Different targeted proteomics strategies have recently emerged in the field of proteomics that enable the detection and quantification of a predetermined subset of proteins with a high degree of sensitivity and reproducibility across many samples. This EMBO Practical Course aims to fill in the gap between theory and the actual implementation of the targeted proteomics workflow, so that by the end of the week the students have the necessary know-how to implement the targeted proteomics workflows in their own research laboratories. The course will offer a daily keynote talk by a high-profile speaker introducing the topic of the day with examples of his/her own research, followed by "Practical demonstrations" (20%), and "Practical work and exercises" (40%) that will cover the complete workflow for experimental design and data analysis of targeted proteomics assays (i.e. targeted method design, optimization of instrument settings, manual and automated quantitative analysis, and statistical analysis of the data). Moreover, these sessions will be followed by "Poster Sessions" (10%) and "Social Events" to introduce informal discussion tables to adapt the technology to each participant's particular interests.

Phosphoinosites are now well recognised as signaling intermediates and their role in cell surface signal transduction and sub-cellular processes such as membrane trafficking have been described. However, in recent years, the availability of high-resolution imaging tools and novel technologies have resulted in new models on the dynamics of PIs in living cellular organelles and the mechanisms by which their levels and distribution are controlled. This in turn has led to emerging ideas on novel modes by which PIs regulate cell architecture and physiology. In parallel, high-throughput genomics technologies have led to the description of genetic changes in protein components of the PI signaling system linked to human disease. Finally, lipidomics technologies are beginning to provide new insights into the acyl chain diversity of PIs that is likely to inform on their function as specific sub-cellular processes. This workshop offers an exciting and opportune moment to discuss the manner in which PIs act as master regulators of cellular architecture and physiology in health and disease.

Recent advances in genomics and proteomics technologies have transformed our ability to study diverse molecular processes. We can now use these technologies to study and understand mechanisms regulating transcription, translation, protein abundance, protein-protein, protein-metabolites and protein-drug interactions at a genome- and proteome-wide scale. The current challenge is to convert this data into predictive quantitative models of cellular processes. This massive task requires intensive integration of basic biology, genomics, proteomics, imaging, computational biology, mathematics, engineering and physics. This conference brings together scientists from diverse fields to promote cross-disciplinary collaborations and form a synthesis of appropriate systems-level approaches. We aim to get people out of their comfort zones, to hear new cutting-edge research in diverse areas to form new ideas. Over the past decade, this conference has served as an important venue to help shape the field and to generate a community of scientists that come from very diverse disciplines, each with the common goal to understand the systems level properties of their system of interest.

Since Wnt research covers various fields of the life science, this meeting is aiming to cover “molecular and cellular biology of the signaling”, “structural biology”, and “stem cell biology”, as well as “clinical application”. An important objective of this EMBO Workshop on Wnt 2022 is to bring together researchers from different fields for exchanging cutting-edge information that advance this research field.

This Workshop will focus on the biology of toxic metabolites in the ageing process, stem cells and cancer. Specific emphasis will be placed on how metabolites that carry carbonyls (such as aldehydes and ketones) damage stem and cancer cells. A particular area will focus on endogenous aldehydes (such as acteladehyde, formaldehyde and methyl-gyoxal), the enzymes that remove them [aldehyde dehydrogenases (Aldh) and functionally related glutathione-based enzymes such as Adh5 and Glo-1/2]. Further, we will consider how human genetic variation can contribute to metabolite sensitivity, and how mutations in cancer genomes in toxic metabolism pathways can be exploited for therapy.

Rapid advancement in the field of NMR spectroscopy in recent years has allowed for the development of novel methods for characterizing biomolecular structure and dynamics. The unique ability of NMR to probe biomolecules under native conditions and near physiological state has enabled researchers to address new problems in biology. This India | EMBO Lecture Course will focus on emerging developments in NMR that will spur further innovation in the areas of protein dynamics, membrane proteins, intrinsically disordered proteins, phase separation, paramagnetic proteins, and large protein complexes. We aim to bring together researchers with diverse interests to realize the full potential of the latest advances in solution and solid-state biomolecular NMR spectroscopy, and to engage early career researchers and leaders in the field in discussions about remaining challenges and the new methods and techniques being developed to address these problems. Bringing together scientists with expertise in different dimensions of NMR will enable facile exchange of ideas, spur collaborative research, and drive future breakthroughs.

In the last decade accumulating evidence have demonstrated that the 20S proteasome can function in a free, uncapped form, leading to the processing and degradation of partially unfolded proteins. Various functional roles have been associated with this activity, including neuronal communication, antigenic peptide production, regulation of protein function through post-translational processing and extracellular activities. However, up until now a community effort to advance this field of research or a platform for exchanging knowledge and ideas was lacking. Therefore this meeting will be specifically dedicated to the independent activity and regulation of the 20S proteasome. We believe that such an event will lead to a supportive and collaborative community and advancement of the research. During the meeting, there will be social events and round-table discussions generating a fruitful platform for establishing international ties and novel collaborations.

This will be the 4th Conference on Bacterial Cell Biology. In the last two decades there has been an explosion of interest in fundamental aspects of bacterial structure and function, based on advances in fluorescence and cryo-EM imaging. Quantitative and automated methods have brought about radical changes in the breadth and depth of questions that could be explored, while genome sequencing and genetic methods such as CRISPR-Cas9 and Tn-Seq have greatly expanded the range of organisms that can be studied incisively, enabling comparative studies of processes such as growth, morphogenesis and division, across diverse bacterial phyla. Many of these bacterial cell biology systems are being successfully reconstituted using purified components, leading to the discovery of molecular mechanisms underpinning cellular functions. The conference will bring together presentations illustrating the application of advanced cell and molecular biology methods to a wide range of basic functions in bacterial cells. Advances in understanding of the fundamentals of bacterial structure and function also create opportunities for the development of novel approaches to kill or modulate the growth and behaviour of bacterial cells.

While alterations to DNA underlie genome evolution and antibody diversification, genome instability drives many disorders, including cancer, developmental abnormalities, and neurodegeneration. DNA damage and other genome-threatening replication blocks are generally overcome by a number of DNA repair pathways. The choice of DNA repair pathways, specifically the reliance on error-prone vs. error-free is both intriguing and confounding. Moreover, the deregulation of repair pathway choice can trigger pathological processes. In this conference, we will discuss the delicate equilibrium of genome maintenance pathways, addressing how natural processes such as replication and transcription can threaten genome stability, how the genome can be altered in cancer, signatures of genome instability in human disease, and how cells control the inherent plasticity of their replication and repair machinery. We aim to bring together scientists from diverse fields who are interested in understanding the many processes impacting DNA metabolism and the molecular and biophysical principles that maintain the balance between genome stability and plasticity.

Meiosis is a defining feature of sexual reproduction. This specialized cell division program comprises a single round of DNA replication followed by two successive cell divisions, thereby reducing chromosome number by half. This reduction accommodates the later combination of two haploid gametes during fertilization to reconstitute a full diploid genome. The unique events of meiosis, including pairwise alignment of parental chromosomes, inter-homolog recombination, kinetochore attachment, cell cycle adaptations and error correction mechanisms, are intensely studied processes in numerous model organisms (fungi, protists, plants, and animals). The Meiosis field is driven by fascination in how evolution has implemented and perfected the meiotic program, and also by its significance to both plant breeding and human health. As a special topic for the 2021 EMBO Workshop, we have chosen homolog pairing. The recognition and physical pairing of homologous chromosomes is an essential feature of meiosis, and also occurs in other contexts. It is therefore timely to bring together the fields of meiotic and post-meiotic chromosome pairing to potentially uncover common principles and mechanisms.

Yet, the known biological mechanisms for inducing changes in the synaptic connections between neurons are driven by neural events that are separated by typically no more than a few tens or hundreds of milliseconds. This conference will convene scientists studying the molecular, cellular, and circuit-level mechanisms used by the brain to bridge the multiple timescales across which events play out in the experience of an individual. The goal will be to assemble an up-to-date understanding of the set of biological mechanisms available in the brain to support associative learning across different timescales. The conference's emphasis on biological mechanisms will complement and support the main program’s more computational focus, and encourage an integrated analysis of statistical learning spanning molecular, cellular, circuit, and behavioral-level phenomena.

Mitochondria act at the interface of numerous processes critical for cellular function and organismal health. Long recognized as the powerhouses of the cell, mitochondria are emerging as hubs for cellular metabolism, redox balance, signalling, gene expression, detoxification, and ion homeostasis. This conference will focus on three major areas: First, it will present new approaches to generate precise and complex genetic alterations in mice and other animals to better “humanize” models and allow for in vivo studies of cancer gene function. Second, it will highlight studies to investigate fundamental mechanisms of cancer initiation and progression, including the cell of origin of tumors, their evolution, their metabolism, and their interactions with the immune system. Finally, the meeting will explore how genetically engineered animal models can help evaluate the efficacy of novel therapeutic approaches and to decipher mechanisms of therapeutic resistance.

The germline provides an eternal cellular link between generations and is the source of genetic diversity and evolution of animal populations. In addition, perturbations in the epigenome or genome of a parental germline that arise during development could be underlying causes of many disease states in offspring. In males, sperm are the conduit for transmission of genetic and epigenetic information to the next generation and are formed by the complex process of spermatogenesis, which involves undifferentiated stem cell, differentiating, meiotic, and morphogenesis phases. This workshop will explore recent advances in understanding the biology of these different phases in a variety of animal models as well as how epigenetic and genetic abnormalities may underlie transmission of diseases and fertility disorder in humans. Invited talks and poster sessions that focus on cutting edge findings along with vibrant discussions and debates about controversies in the field will provide a rich intellectual atmosphere that is sure to have an important impact on how the biology of spermatogenesis is investigated for years to come.

The maintenance of genome integrity is critical for the suppression of several pathological disorders in humans, including cancer, infertility and neurodegeneration. Moreover, the accumulation of unrepaired errors in DNA is commonly cited as a likely cause of tissue and organismal ageing. Destabilization of the genome can occur as a result of several cell intrinsic or extrinsic factors, including errors arising during DNA replication or chromosome segregation, as well as exposure of cells to agents that induce DNA damage. In this conference, we aim to bring together scientists studying DNA replication, DNA repair and chromosome segregation with those interested in how chromosomal instability can influence human pathology. Moreover, we aim to show how high throughput and high content screening methods can be used as a discovery tool both for basic science applications and to identify potential therapeutic modalities.
