The RESOLUTE and REsolution consortia of the Innovative Medicines Initiative of the European Union have been empowering over the last years the scientific community with novel research tools and datasets for transporters of the Solute Carrier (SLC) supergroup. In 2023, both projects are coming to an end, and we would like to finish with a big celebratory and future-looking conference: “Unlocking Transporters for Drug Discovery”. At the event expect to see presented the results and lessons learned by the RESOLUTE and REsolution consortia over the last five years. Furthermore, top international scientists interested in metabolism and its regulation, drug discovery, the function and structure of membrane proteins will be speaking.

For decades, studies in cell biology have focused on discovering the functions and dynamics of individual organelles. However, organelles do not exist only as single units, but also as part of entities formed through contacts between two or more distinct organelles. In fact, the recent years of studies on inter-organelle contacts have revealed that complex cellular functions, biochemical processes and signaling pathways are governed at the sites where heterotypic organelles come into contact with each other. This paradigm shift has spurred a new gold rush of research in cell biology that is transforming our understanding of cell physiology, signaling and metabolism in health and disease. This EMBO Workshop on inter-organelle contacts biology, now at its 3rd edition, aims at bringing together the community of scientists in this field to share their work, experience, expertise, and vision.

The germline provides an eternal cellular link between generations and is the source of genetic diversity and evolution of animal populations. In addition, perturbations in the epigenome or genome of a parental germline that arise during development could be underlying causes of many disease states in offspring. In males, sperm are the conduit for transmission of genetic and epigenetic information to the next generation and are formed by the complex process of spermatogenesis, which involves undifferentiated stem cell, differentiating, meiotic, and morphogenesis phases. This workshop will explore recent advances in understanding the biology of these different phases in a variety of animal models as well as how epigenetic and genetic abnormalities may underlie transmission of diseases and fertility disorder in humans. Invited talks and poster sessions that focus on cutting edge findings along with vibrant discussions and debates about controversies in the field will provide a rich intellectual atmosphere that is sure to have an important impact on how the biology of spermatogenesis is investigated for years to come.

The maintenance of genome integrity is critical for the suppression of several pathological disorders in humans, including cancer, infertility and neurodegeneration. Moreover, the accumulation of unrepaired errors in DNA is commonly cited as a likely cause of tissue and organismal ageing. Destabilization of the genome can occur as a result of several cell intrinsic or extrinsic factors, including errors arising during DNA replication or chromosome segregation, as well as exposure of cells to agents that induce DNA damage. In this conference, we aim to bring together scientists studying DNA replication, DNA repair and chromosome segregation with those interested in how chromosomal instability can influence human pathology. Moreover, we aim to show how high throughput and high content screening methods can be used as a discovery tool both for basic science applications and to identify potential therapeutic modalities.

Over the past decades it became evident that a RNA transcripts specific amount and localisation are important in order to understand the biological function of its (non-) coding gene. Whereby state-of-the-art in situ hybridisation (ISH) technologies are key to visualise these RNAs in a given tissue or cell. This course aims to highlight three powerful RNA visualisation techniques in histological tissue samples. During this course, the participants will learn to validate the cellular localisation of candidate genes from transcriptome studies through conventional ISH and, the more recent cutting-edge, smFISH (Single Molecule Fluorescent ISH) and HCR-ISH (Hybridization Chain Reaction), based RNA ISH hybridisation methods. Additionally, the course will also cover essential tissue embedding protocols that are adapted for use in visually interrogating RNA transcripts in mammalian tissues.

This course consists of two parts: virtual and onsite. The virtual course takes place 6 – 10 November 2023 and the onsite part takes place 27 November – 1 December 2023.

The virtual course will cover the basics of SAXS/SANS, instrumentation, data collection, modelling techniques and complementary use with other methods. The students are encouraged to sign-up for a self-paced practical tutorial that covers basic and advanced SAXS data processing steps (including quizzes to keep track of successful participation). In addition, students are encouraged to apply to the ‘add on module’ which includes an on-site visit (5 days) in Hamburg to participate either in track 1: X-ray beamline operation or track 2: sample optimisation for SAXS. Students not travelling to Hamburg can sign up for small discussion groups to discuss their SAXS data in more detail.

Flow Cytometry is one of the most powerful single cell analysis methods used throughout the life sciences and medicine. By utilising Flow Cytometry we can identify and quantify specific cell populations from heterogeneous samples, analyse the cell cycle or proliferation state or simply detect the expression of fluorescent reporters. In line with the suggestions from our course in 2019, we will significantly enhance the number of analysis sessions together with our partners from FlowJo. We will cover the theory of Flow Cytometry and corroborate this information intensively with the practical issues of the most frequent assays as part of our practical course curriculum.

The safety, quality and clinical results of drug substances and drug products may be disrupted by the formation of nitrosamines along with other mutagenic impurities. The comprehensive development and production of innovative medicines now inevitably involves estimating impurity profiles as well as managing and monitoring them. To meet the demands of ever-increasing regulatory standards and avoid expensive recalls, proactive detection of genotoxic contaminants should be adopted at every stage of pharmaceutical development. We plan an online meeting with subject-matter experts to evaluate multiple perspectives on how to handle the concerns of mutagenic impurities. We will cover regulatory-compliant drug impurity profiling practices; helpful hints for adhering to recent ICH M7 and Q3D updates; nitrosamine impurity identification and quantification; risk evaluation of genotoxic impurities; analytical challenges and genotoxicity prediction; establishment of acceptance criteria; and the most recent developments in the analysis of elemental impurities. The research field of interorganelle contacts is one of the most rapidly growing areas of modern cell biology. It is becoming clear that many, if not all, organelles form membrane contact sites (MCS) with other components of the cell. The term ‘MCS’ broadly describes a physical interaction between two organelles, occurring at specialized subdomains of the given organelle and involved in multiple facets of the biology of the two organelles that are juxtaposed. MCS discovery forced cell biologists to reconsider the way organelles are studied, introducing the concept that heterotypic organelle interaction allows functions that individual organelles would otherwise be unable to perform and forcing them to think in terms of compartmentalized metabolism and signaling.

Evolutionary cell biology represents the integration of evolutionary theory, phylogenetics and experimental evolution with comparative cell biology, aiming ultimately to understand how and why cells evolved to be the way they are. This meeting aims to bring together evolutionary biologists and cell biologists investigating diverse aspects of cellular physiology, traversing both spatial scales – from subcellular cytoskeletal dynamics to symbiotic interactions – and evolutionary time. We have reached a fascinating point in our research on the ubiquitin system’s function, physiology and roles in disease. The attachment of ubiquitin to a protein in the cytosol was first identified as a means to enable degradation of proteins in the cytosol by the proteasome. It was much later that it was discovered that the ubiquitin system fulfils another crucial cellular function: through the dynamics offered by ubiquitination and subsequent deubiquitination it gives a multitude of signalling pathways the required plasticity to initiate but also to limit their signalling output in an exquisitely controlled manner. Whereas certain signalling-related functions employ ubiquitin’s proteasome-targeting function, e.g. the activation of NF-kB following degradation of I-kB, most signalling-related functions of the ubiquitin system are mediated by different types of ubiquitin chains acting as scaffolds for the recruitment of signalling modules to protein complexes which serve as platforms for the triggering of specific signalling complexes.

Likely pathways from the inanimate to the animate world involve a transition that can be thought of as the emergence of information in molecular systems. Before this transition, heteropolymers already exist, but the sequence information of a polymer does not persist beyond the lifetime of the polymer. During the transition, information becomes a separate dynamical entity, which lives and changes over longer timescales than its carrier molecules. This transition does not necessarily require the existence of a full-fledged replication machinery, such as polymerase enzymes that copy informational polymers. Instead, the interplay of simpler reactions can create strong correlations in pools of informational polymers, which can also decouple the timescales of the informational dynamics from those of the polymer dynamics. This interplay is only beginning to be studied, experimentally as well as computationally and theoretically, such that many conceptual and methodological questions are currently open. The central aim of this workshop is to bring together scientists with different backgrounds, in order to attack these questions together.

We will highlight critical issues on lung epithelial biology such as stem cells and regenerative medicine, while expanding sessions to encompass themes such as single cell analytical approaches, cell-cell communication, dissect the biological distinctions mesenchymal proliferation as a repair mechanism and fibrosis; present the impact of new tools in single cell analysis on understanding lung development, repair, and disease; and characterize epithelial-mesenchymal interrelationships that maintain lung homeostasis and orchestrate repair and regeneration. The entire program has been designed to achieve a better understanding of the key clinical research issues and how they relate to basic mechanistic investigation.

Defects in DNA damage repair and checkpoint control are underlying mechanisms driving tumorigenesis, since they allow for the accumulation of genetic alternations. Indeed, defects associated with these pathways are the underlying cause of several human cancer-prone syndromes. On the other hand, defects in DNA repair and checkpoint control create vulnerabilities in cancer cells that can be targeted by DNA damaging agents and inhibitors that disrupt checkpoint pathways. More recently, defective DNA repair and checkpoint regulation have been shown to influence R-loop formation, replication fork stability, and innate immunity. These new findings not only reveal additional mechanisms contributing to tumorigenesis, but also provide new targets for cancer therapy. This conference will focus on exploring these vulnerabilities, taking advantage of the synthetic lethality concept, and targeting specific DNA repair and checkpoint pathways for cancer therapy, either alone or in combination with chemotherapy, radiation therapy, targeted therapy, and immunotherapy. This conference will bring together basic, translational and clinical investigators to discuss current and future directions, opportunities and obstacles to the development of these anti-cancer modalities and how to best translate these concepts to clinical practice.

Chromatin is the template of life, the carrier of genetic information and the core regulator of fundamental processes such as gene expression and stem cell fate determination. Chromatin dysfunction on the other hand can trigger various diseases, including cancer. Although chromatin structure and function and the mechanisms of disease and their diagnostic and therapeutic approaches are the subject of many meetings, conferences bringing them together are less frequent. The boundaries among these various disciplines are disappearing and meeting on "Epigenetics: from mechanisms to disease" will focus on the discussion of the role of chromatin architecture, epigenetics, stem and single cell biology in cancer and other diseases. The talks will cover a broad range of topics, including chromosome organisation, long-range interactions, chromatin assembly, stem cell regulation/differentiation, RNA-based mechanisms, transcription regulation, DNA methylation and hydroxymethylation, exploiting the power of genome engineering technologies, single-cell approaches, cutting edge epigenomics and imaging approaches and novel cell systems such as organoids.
