Abstract:
Light microscopy is the most widely used form of microscopy for life science research. The correct handling of the microscope and optimising image quality (e.g. contrast, resolution) as well as experimental design according to different applications are crucial to obtain the best results. This course will provide the participants with an overview about the basics of microscopy (light and optics, contrast techniques and imaging). The confocal part will include multicolor, 3D and time lapse imaging as well as spectral imaging, photobleaching (FRAP, FLIP, FLAP) and photoactivation techniques. Basic image analysis tools including measurement functions, colocalisation and spectral unmixing as well as 3D/4D image processing will be covered, both in theory and practice. The course will combine lectures with intense daily hands-on practices followed by discussions.

Abstract:
The EMAS Regional Workshops are biennial events designed to provide postgraduate-level and research workers in materials science, material engineering and related subjects, with basic knowledge of the capabilities and limitations of microbeam analysis. The 2018 meeting will focus on the specific requirements of microbeam analysis as utilised by the Earth Science community. The meeting is organised in collaboration with the Mineralogical Society of GB and Ireland who presented the workshop “Microbeam Techniques in Geosciences” at Imperial College, London in 1998. Here we revisit the subject to look at the range of techniques now available and the technological advances made in the intervening two decades.

The focus of the course will be on the use of fluorescence microscopy to obtain quantitative information about protein dynamics in living samples.

Abstract:
Cryo-EM combined with image processing is currently undergoing what has been referred to as a “resolution revolution”, making it one of the most powerful techniques to address this challenge. It also has unique capabilities to obtain structural information in situ within complex environments, and to deal with heterogeneous samples. The methods are complex and under constant development, so in depth, advanced training of users is essential to ensure future users can adapt new technologies to expand into new areas and solve new biological problems. This course provides the theoretical and practical training covering the full breadth of cryo-electron microscopy (cryo-EM) techniques in order develop novel image processing approaches.

EMBL Course: Gene Expression at Spatial Resolution

Abstract:
This course will introduce participants to a new generation of a spatially resolved transcriptomics assay provided by 10x Genomics. Through the combination of state-of-the-art sequencing technology and high-resolution microscopy, 10x Genomics is enabling researchers to study gene expression of cells within their morphological context of an intact tissue section. Through both theoretical sessions, as well as extensive hands-on practical work, the participants will learn the entire workflow of sample preparation including cryosectioning of fresh-frozen tissue samples, optimisation of tissue fixation and permeabilisation, high-resolution microscopy and sequencing library preparation. Utilising the 10x Genomics Loupe Browser pipeline, the participants will also have hands-on training of spatial transcriptomics data analysis.

These revolutionary developments come with novel requirements for fluorescent probes, labelling techniques, and data interpretation strategies. Researchers from across the scientific spectrum are involved in designing improved tools and solving previously intractable problems using super-resolution techniques. We invite you to attend the ‘International Conference On Nanoscopy 2020’, which will uniquely and exclusively focus on super-resolution microscopy and its biological application. Held in Jena, Germany in the summer of 2020, it will bring together an international group of experts to discuss the latest advances and future directions in the field.
