Presentation — Micro-characterisation Applications of Cultural Artifacts

Abstract:
The application of advanced micro and nano-scale characterisation techniques is now popular in the study of cultural heritage artifacts. This presentation will highlight a number of such applications from the wide range of collaborations with non-profit institutions in south eastern Michigan. Analyses were preformed by optical microscopy, transmission and scanning transmission electron microscopes and environmental scanning electron microscopy.

This practical course will cover advanced light microscopy techniques and participants will learn how to derive qualitative and quantitative insights on molecular mechanisms in cells and developing organisms. Invited guest researchers together with microscopy experts from the EMBL will foster an intense information flow with a balance of lectures and practical workshops. The focus of the course will be on the use of fluorescence microscopy to obtain information about localisation and functions of proteins and other bio-molecules at different spatial scales.

Abstract:
Expansion microscopy (ExM) physically magnifies specimens, allowing to obtain super-resolution images using a conventional diffraction-limited microscope such as confocal microscopy. This powerful technique is easy to implement but it requires some hands-on training. This workshop aims at disseminating and discussing expansion microscopy protocols and is intended for cell biologists, parasitologists, etc, with applications ranging from tissue to cells.

The main goal of this course is to practically demonstrate a broad variety of SRLM methods. During the course, a majority of available SRLM systems will be demonstrated and available for hands-on experience. Leading experts of the field will present their research and methodology. Many opportunities for discussions and social networking will provide a unique and highly efficient environment to support sharing of skills and knowledge. This promotes establishing of contacts and communication channels within the in-field scientific SR community. The focus of the course is on the practical part with a lot of hands-on sessions supported by theoretical lectures and complemented by presentations of ongoing scientific projects presented by leaders of the field.

Abstract:
Light microscopy is the most widely used form of microscopy for life science research. The correct handling of the microscope and optimising image quality (e.g. contrast, resolution) as well as experimental design according to different applications are crucial to obtain the best results. This course will provide the participants with an overview about the basics of microscopy (light and optics, contrast techniques and imaging). The confocal part will include multicolor, 3D and time lapse imaging as well as spectral imaging, photobleaching (FRAP, FLIP, FLAP) and photoactivation techniques. Basic image analysis tools including measurement functions, colocalisation and spectral unmixing as well as 3D/4D image processing will be covered, both in theory and practice. The course will combine lectures with intense daily hands-on practices followed by discussions.
