Emerging Theoretical Approaches to Complement Single-Particle Cryo-Electron Microscopy

Abstract:
The field of biology is experiencing a transformative phase, thanks to remarkable advances in single-particle cryo-electron microscopy (cryo-EM). The explosion of high-resolution cryo-EM maps is challenging computational biophysics to rapidly and robustly discern mechanistic implications from the experimental data. Emerging theoretical approaches aim at processing, validating, complementing, and interpreting cryo-EM data, overcoming issues in image analysis, map refinement, and the simulation of large biomolecules. This thematic meeting seeks to explore the interface between computational biophysics and cryo-EM, highlighting the breadth of work that spans these two fields, and encouraging new synergies. Our goal for this meeting is to maximize the potential of computations and experiments in the field of single-particle cryo-EM.

EIC24-01 — Imaging down to single-molecule resolution: STED & MINFLUX nanoscopy

Abstract:
The course aims to train biological microscopy users in two forms of super-resolution light microscopy, Stimulated Emission Depletion (STED) Microscopy and Minimal photon fluxes (MINFLUX). Both techniques require technical understanding of the process and instrumentation, training in the method specific requirements of sample preparation and in the acquisition procedures and an understanding of how to further analyze the acquired image data. It is the aim of the course to provide the tools for the successful execution of nanoscopy experiments with these two distinct methods. The course should also allow the participants to understand the specific benefits of the different techniques and thus help them in choosing the right approach for their experimental needs.

The NEMI infrastructure consists of a distributed network of local EM Nodes and a number of unique Flagship Nodes, which offer highly advanced EM technologies requiring specific expertise and often very costly instrumentation. Together, these technologies enable imaging across different length, resolution and time scales and allow integration with light microscopy and proteomics.

This meeting will consist of themed plenary sessions with talks from invited speakers. There will also be parallel scientific workshops organised by members of the cytometry community and parallel commercial workshops. There will be a large exhibition and the opportunity to network with flow and image cytometrists from all over Europe and beyond. The meeting will highlight advances in flow and image instrumentation, high content screening, cancer and stem cell biology, applications of clinical cytometry and the development of novel probes and approaches in many areas of biomedical research.

Abstract:
Microscopy is a key methodology in life science research, where the reliability of scientific findings based on data generated on microscopes rely on substantiated qualitative and quantitative image data analysis. Due to the complexity of the data, without expert guidance, it is very likely that image analysis algorithms may be applied incorrectly, possibly even producing erroneous results. Moreover, the diversity of imaging modalities, analysis algorithms and software solutions is growing so rapidly that even experts are overwhelmed. This course aims to promote excellence in bioimage analysis, targeting scientists who already work in this area or harbor ambitions to do so – and will instruct on the latest developments in the field with ample group and networking activities with leading experts in the topic.
